摘 要:目的:比较不同提取溶剂对智脑胶囊药材的主要成分含量的影响,为智脑胶囊的提取工艺选择提供依据。方法:按处方取智脑胶囊药材饮片共20份,分别以水和75%乙醇作为溶剂提取,各10批;采用UPLC法测定松果菊苷、毛蕊花糖苷、毛蕊异黄酮苷、党参炔苷、槲皮素指标成分的含量,建立其UPLC指纹图谱。色谱柱为Waters Acquity BEH C18色谱柱(100 mm×2.1 mm,1.7μm),流动相为乙腈(A)-0.05%甲酸溶液(B),梯度程序洗脱,流速为0.20 mL/min;柱温为30℃,检测波长为260 nm;分别采用指标成分含量、指纹图谱相似度评价、聚类分析和OPLS-DA分析对水提、醇提工艺提取效果进行比较。结果:建立的5种指标成分UPLC测定和指纹图谱方法可行;智脑方醇提工艺样品溶液中5种成分含量较水提工艺样品高3.52%~130.73%;75%乙醇提取的样品指纹图谱色谱峰数较水提样品明显增多,水提、醇提工艺样品各自的相似度较好,聚类分析和OPLS-DA分析可以明显地区分2种工艺样品。结论:以水和75%乙醇作为提取溶剂制备的智脑胶囊溶液,其主要成分含量和指纹图谱存在较明显差异,以75%乙醇为溶剂提取的效果更好。Objective:To compare the effects of different extraction solvents on the main component content of Zhinao Capsules,and provide a basis for the selection of extraction technology for Zhinao Capsules.Methods:According to the prescription,20 pieces of medicinal materials of Zhinao Capsule were extracted with water and 75%ethanol as solvent,10 batches each.The UPLC method was used to determine the contents of echinacin,verbascum thapsus glycoside,piloside Isoflavone glycoside,codonosyl glycoside and quercetin,and establish their UPLC fingerprints.The chromatographic column was Waters Acquisition BEH C18 chromatographic column(100 mm×2.1 mm,1.7μm),the mobile phase was acetonitrile(A)-0.05%formic acid solution(B),eluted by gradient program,with a flow rate of 0.20 mL/min.The column temperature was 30℃,and the detection wavelength was 260 nm.The results of aqueous extraction and alcohol extraction were compared by indicator component content,fingerprint similarity evaluation,cluster analysis and OPLS-DA analysis.Result:The established UPLC determination and fingerprint methods for the 5 indicator components were feasible.The content of five components in the sample solution of Zhinao Capsule alcohol extraction process was 3.52%-130.73%higher than that of the aqueous extraction process sample.The number of chromatographic peaks in the fingerprint spectrum of samples extracted with 75%ethanol was significantly higher than that of water extracted samples.The similarity between water extracted and alcohol extracted process samples was good,and cluster analysis and OPLS-DA analysis can clearly distinguish the two process samples.Conclusion:The main components and fingerprint of Zhinao Capsule solution prepared with water and 75%ethanol as the extraction solvent are significantly different,and 75%ethanol as the solvent has better extraction effect.
