摘  要:目的:以抗炎有效成分为评价指标,建立畲药树参茎的质量标准。方法:采用显微鉴别法对树参茎粉末的特征性组织、细胞及细胞内含物进行鉴别;采用薄层色谱法对树参茎中的4个主要抗炎有效成分(紫丁香苷、绿原酸、异绿原酸A、异绿原酸C)进行鉴别;采用高效液相色谱法建立树参茎中上述4种成分的含量测定方法;并对树参茎的水分、总灰分、浸出物进行控制。结果:建立的显微鉴别法可鉴别出树参茎粉末中的木纤维、木栓组织碎片细胞、草酸钙簇晶、石细胞、导管、韧皮纤维;建立的两个薄层色谱法,可分别鉴别出树参茎中的主要抗炎有效成分紫丁香苷、绿原酸、异绿原酸A和异绿原酸C;建立的两个高效液相色谱法,可分别对树参茎中的紫丁香苷、绿原酸、异绿原酸A、异绿原酸C进行含量测定,4种成分的线性范围分别为0.0357~0.8940μg、0.0775~1.9380μg、0.0223~1.1140μg、0.0123~0.6170μg;回归方程分别为y=1829.1x+3.5032(r=0.9998)、y=531.13x-5.3732(r=0.9998)、y=3296.4x+0.3842(r=0.9999)、y=3493.8x-13.2120(r=0.9999);平均加样回收率分别为98.52%、99.83%、98.32%、99.42%,RSD分别为0.57%、0.57%、0.86%、0.96%。结论:建立的方法简便、准确、重现性好,以抗炎有效成分为评价指标建立的质量标准更有利于发挥树参茎的临床疗效。Objective:To establish the quality standard of the stems of She Medcine——Dendropanax dentiger with the anti-inflammatory active ingredients as the evaluation index.Methods:The characteristic tissues,cells and cell contents of the stem powders of Dendropanax dentiger were identified by microscopic identification.Thin layer chromatography(TLC)was used to identify the four main anti-inflammatory active ingredients in the stems of Dendropanax dentiger,including syringin,chlorogenic acid,isochlorogenic acid A and isochlorogenic acid C.High perfromance liquid chromatography(HPLC)was established for the determination of four main anti-inflammatory active ingredients.And we controlled the water content,total ash and extract of the stems of Dendropanax dentiger.Results:The established microscopic identification could identify wood fibers,cork tissue fragment cells,calcium oxalate cluster crystals,stone cells,vessels and phloem fibers in the stem powders of Dendropanax dentiger.There were two TLC methods established to identify syringin,chlorogenic acid,isochlorogenic acid A and isochlorogenic acid C.There were two HPLC methods established to determine the contents of the above active ingredients.The linear ranges of the four components were respectively 0.0357-0.8940μg,0.0775-1.9380μg,0.0223-1.1140μg and 0.0123-0.6170μg.And the calibration curve was respectively y=1829.1x+3.5032(r=0.9998),y=531.13x-5.3732(r=0.9998),y=3296.4x+0.3842(r=0.9999)and y=3493.8x-13.2120(r=0.9999).The average recovery rate was respectively 98.52%(RSD=0.57%),99.83%(RSD=0.57%),98.32%(RSD=0.86%)and 99.42%(RSD=0.96%).Conclusion:The established method is simple,accurate and reproducible.The quality standard established with anti-inflammatory active ingredients as the evaluation index is more conducive to the clinical efficacy of the stems of She Medcine-Dendropanax dentiger.
