摘  要:目的探讨参附注射液对慢性心力衰竭大鼠模型的保护作用以及Toll-like受体(TLRs)信号通路在其中的作用。方法构建大鼠心衰模型,并随机将40只大鼠分为对照组、参附注射液低剂量组、参附注射液高剂量组和模型组。使用心肌细胞分别进行了实时定量PCR和Westernblot分析,检测Toll-like受体4(TLR4)、髓样分化因子(MyD88)、肿瘤坏死因子受体相关分子6(TRAF6)的mRNA和蛋白表达。结果参附注射液高剂量组中,TLR4、MyD88和TRAF6的mRNA表达水平明显下降(低剂量组降低不显著)。参附注射液高剂量组中,TLR4、MyD88、TRAF6蛋白表达水平显著下降(低剂量组降低不显著)。结论参附注射液在慢性心力衰竭大鼠模型中有保护心肌作用,它可能是通过抑制炎症反应以及心肌纤维化来实现的,其机制与TLRs信号路径有关。Objective To investigate the protective effect of Shenfu injection on chronic heart failure rat model and the role of Toll-like receptor(TLRs)signaling pathway.Methods A rat heart failure model was established,and 40 rats were randomly divided into control group,Shenfu injection low-dose group,Shenfu injection high-dose group and model group.The mRNA and protein expressions of Toll-like receptor 4(TLR4),myeloid differentiation factor(MyD88)and tumor necrosis factor receptor-associated molecule 6(TRAF6)were detected by real-time quantitative PCR and Western blot analysis of cardiomyocytes,respectively.Results The mRNA expression levels of TLR4,MyD88 and TRAF6 in the high-dose group of Shenfu injection were significantly decreased(the decrease was not significant in the low-dose group).The protein expression levels of TLR4,MyD88 and TRAF6 in the high-dose group of Shenfu Injection were significantly decreased(the decrease was not significant in the low-dose group).Conclusion Shenfu injection can protect myocardium in rats with chronic heart failure,which may be achieved by inhibiting inflammatory response and myocardial fibrosis,and its mechanism is related to TLRs signaling pathway.
