摘  要:【目的】探究NtJAZ1敲除对烟草响应外源茉莉酮酸甲酯(methyl jasmonate,MeJA)对生长发育、腺毛发生、烟碱生物合成的应答反应的影响。【方法】以野生型K326为对照,比较不同浓度MeJA处理下,NtJAZ1敲除的K326突变品系NK-9在种子萌发、根系发育、腺毛形态、烟碱生物合成等方面的差异。【结果】(1)MeJA处理显著抑制了种子的萌发,在100μmol/L MeJA处理条件下,K326的种子萌发率比未经MeJA处理的种子萌发率降低了85.72%,NK-9的种子萌发率比未经MeJA处理的降低了48.12%。(2)MeJA处理抑制根系发育,在300μmol/L MeJA处理条件下,K326幼苗主根比未经处理的缩短了83.33%,NK-9幼苗主根缩短了68.18%。(3)MeJA处理诱导腺毛发生,在5 mmol/L MeJA处理条件下,K326叶面长柄腺毛密度比未经处理的提高了42.55%,NK-9提高了21.05%。(4)MeJA处理促进烟碱生物合成,在5 mmol/L MeJA处理条件下,K326烟苗的烟碱含量比未经处理的提高了76.05%,NK-9提高了37.51%。【结论】NtJAZ1敲除影响了烟草对MeJA的应答反应,减缓了MeJA对种子萌发与根系发育的抑制作用,也减缓了MeJA对烟草叶面长柄腺毛发生和烟碱生物合成的促进作用。[Objective]This study aims to investigate the effects of NtJAZ1 knockout on the response of tobacco to exogenous methyl Jasmonate(MeJA)as well as on glandular trichome development,and nicotine synthesis of tobacco.[Methods]In this study,NK-9,the NtJAZ1-knockout K326 mutant,was used to analyze seed germination,root development,glandular trichome morphology,and nicotine biosynthesis under different concentrations of MeJA treatment.[Results]1)MeJA treatment significantly inhibited seed germination.The germination rates of NK-9 and K326 decreased by 48.12%and 85.72%under 100μmol/L MeJA treatment,respectively.2)MeJA treatment significantly repressed root development.The taproots of NK-9 and K326 seedlings were shortened by 68.18%and 83.33%under 300μmol/L MeJA,respectively.3)MeJA treatment induced the development of long stalk glandular trichomes.Under 5 mmol/L MeJA treatment,the density of long stalk glandular trichomes of NK-9 and K326 increased by 21.05%and 42.55%,respectively.4)MeJA treatment modulated nicotine synthesis.Nicotine accumulation in shoots of NK-9 and k326 increased by 37.51%and 76.05%under the treatment of 5 mmol/L MeJA,respectively.[Conclusion]The results showed that NtJAZ1 knockout affected the response of tobacco plants to MeJA,and slowed down the inhibitory effects of MeJA on seed germination and root development,and attenuated the promoting effect of MeJA on long stalk glandular trichinosis and nicotine biosynthesis in tobacco.
