摘  要:目的基于生物信息学分析血管抑制蛋白2(VASH2)在乳腺癌(BC)中的表达和临床意义。方法通过癌症基因组图谱(TCGA)联合基因组织表达数据库(GTEx)、基因表达数据库(GEO)及UALCAN检测在BC与正常组织样本中VASH2的表达差异。运用Kaplan-Meier plotter分析VASH2表达水平与患者总生存期的关系。利用UALCAN与cBioPortal协同分析VASH2的表达与甲基化水平的相关性。采用LinkedOmics和Metascape网站进行基因富集分析,研究VASH2在BC发生发展中涉及的分子功能和相关通路。使用TIMER分析VASH2与BC免疫浸润细胞关联性。结果VASH2在BC组织中与正常组相比表达水平显著提高。生存曲线提示高表达VASH2的BC患者总生存期较差。在BC中下调VASH2基因的甲基化修饰程度可能是VASH2表达上调的原因之一。VASH2可能参与细胞周期、BRCA2协作PALB2同源重组修复、有丝分裂G1期和G1/S过渡等信号通路。结论在BC组织中VASH2表达升高且与BC分化程度、淋巴结转移和临床预后相关,可能作为乳腺癌诊断、治疗和预后预测的新分子指标。Objective To analyze the expression and clinical significance of vashohibin-2(VASH2)in breast cancer(BC)based on bioinformatics.Methods Differences in VASH2 expression in BC and normal tissues were detected through The Cancer Genome Atlas(TCGA),Genotype-Tissue Expression(GTEx),Gene Expression Omnibus database(GEO)and UALCAN.The correlation between VASH2 expression and overall survival of patients was analyzed by the Kaplan-Meier plotter.The correlation between VASH2 expression and methylation levels was analyzed using UALCAN in conjunction with cBioPortal.Gene enrichment analysis was performed using LinkedOmics and Metascape websites to investigate the molecular functions and associated pathways involved in the development of VASH2 in BC.In addition,VASH2 was analyzed for association with BC infiltrating immune cells using the Tumor Immune Estimation Resource(TIMER).Results VASH2 expression levels were significantly higher in BC tissues than in normal tissues.The survival curve suggested that BC patients with high VASH2 expression had shorter overall survival.The down-regulation of methylation modification of the VASH2 gene in BC tissues might contribute to the up-regulation of VASH2 expression.VASH2 might be involved in signaling pathways such as cell cycle,homologous recombination repair by breast cancer gene 2(BRCA2)collaborating with partner and localizer of BRCA2(PALB2),as well as mitotic G1 phase and G1/S transition.Conclusion VASH2 expression is elevated in BC tissues and correlated with the degree of BC differentiation,lymph node metastasis and clinical prognosis,and may serve as a new molecular indicator for diagn osis,treatment and prognosis prediction of BC.
