基于ISSR标记的黔西南州光叶子花遗传多样性分析

摘  要:光叶子花具有生命力强、色彩鲜艳、花色繁多、花期长等特点,且在黔西南州分布较广,但同时光叶子花存在不耐寒、易受涝、杂交结实率低等缺点。此次研究运用ISSR分子标记技术,对黔西南州24份光叶子花的遗传多样性进行分析,从26条ISSR通用引物中反复筛选,最终筛选出了6条扩增条带清晰且重复性好的引物,被筛选出来的6条ISSR引物共扩增出24个位点,其中多态性位点23个,PPB为95.833%。根据ISSR群体扩增的结果,进行数据统计后,利用Popgene32软件进行遗传多样性分析,结果显示,Shannon′s信息多样性指数I为0.5710,Nei′s基因多样性指数H为0.3982。根据NTSYS2.1软件对光叶子花种质资源进行聚类分析所获得的树状图显示,遗传相似系数为0.81时,24份光叶子花可被分为2个类群。从H、I和树状图可看出,黔西南州光叶子花具有较高的遗传多样性,此次研究为光叶子花种质资源保护和群体遗传选育研究奠定了遗传学基础。The Bougainvillea glabra Choisy has the characteristics of strong vitality,bright colors,diverse flower colors,and long flowering period,and is widely distributed in Qianxinan Prefecture.However,at the same time,the Bougainvillea glabra Choisy has disadvantages such as low cold resistance,susceptibility to waterlogging,and low hybrid seed setting rate.This study uses ISSR molecular marker technology to analyze the genetic diversity of 24 Bougainvillea glabra Choisy samples in Qianxinan Prefecture.Through repeated screening from 26 ISSR universal primers,6 primers with clear amplification bands and good repeatability are ultimately selected.The 6 ISSR primers selected amplifies a total of 24 loci,including 23 polymorphic loci,with a PPB of 95.833%.Based on the results of ISSR population amplification,data analysis is conducted using Popgene32 software.The results showed that Shannon's information diversity index I is 0.5710,and Nei's gene diversity index H is 0.3982.According to the dendrogram obtained from the clustering analysis of the germplasm resources of Bougainvillea glabra Choisy using the NTSYS2.1 software,when the genetic similarity coefficient is 0.81,24 Bougainvillea glabra Choisy can be divided into 2 groups.From the H,I,and dendrogram,it can be seen that the Bougainvillea glabra Choisy in Qianxinan Prefecture have high genetic diversity.This study has laid a genetic foundation for the protection of germplasm resources and population genetic selection of Bougainvillea glabra Choisy.
