摘  要:北美冬青组织培养技术可以弥补传统繁殖方式无法大量增殖和产业化生产的缺点,能快速生产大量种苗,从而满足市场需求。为建立北美冬青奥斯特组培植株再生体系,以枝条为材料,探讨取材部位、接种方式、生长调节剂等对奥斯特组织培养的影响。结果表明:北美冬青奥斯特的最佳增殖培养基为MS+ZT 4.0 mg/L+IBA 0.2 mg/L,其最高增殖系数为3.29,且芽丛生长旺盛;接种枝条上部茎段新梢诱导率和增殖系数显著低于接种枝条中部和下部,垂直接种效果优于水平接种;最佳生根培养基为1/2 MS+IBA 4.0 mg/L,生根率为44.70%,根系较长且健壮。The tissue culture technology of Ilex verticillata can make up for the shortcomings of traditional repro-duction methods that cannot proliferate in large quantities and achieve industrialization,and it can quickly produce a large number of seedlings to meet market demand.In order to establish a plant regeneration system for tissue culture of Ilex verticillata′Oosterwijk′,this paper took branches as materials,and studied the effects of sampling part,inoculation way and growth regulators on tissue culture of Ilex verticillata′Oosterwijk′.The results showed that the optimum medium for multiplication was MS+ZT 4.0 mg/L+IBA 0.2 mg/L,and the maximum multiplication coefficient was 3.29,and the buds grew vigorously.The new shoot induction rate and propagation coefficient of the upper part of branches were significantly lower than those of the middle and lower parts of the branches,and vertical inoculation was superior to horizontal inoculation.The optimum medium for rooting was 1/2 MS+IBA 4.0 mg/L,the rooting rate reached 44.70%,and the roots were long and robust.
