摘　　要：为了建立蓬莱松组培无菌体系,比较了不同消毒时间、外植体类型、启动和继代培养基配方等的影响,并筛选出了适宜的条件。结果表明,宜选用蓬莱松幼嫩的茎段作外植体,采用0.1%升汞消毒8~<10 min即可;启动培养基配方为MS+6-BA 0.5 mg/L+IBA 1.0 mg/L+蔗糖30 g/L+琼脂4.5 g/L,其腋芽诱导率可达60%;继代培养基最适配方为MS+6-BA 0.6 mg/L+IBA 1.0 mg/L+蔗糖30 g/L+琼脂4.5 g/L,启动培养基上诱导出的芽可在该培养基上正常生长,培养90 d后,可形成丛生芽。组培无菌体系的建立可为后续蓬莱松组培快繁体系的完善提供基础。In order to establish the aseptic system of Asparagus retrofractus in tissue culture,the effects of diffe-rent disinfection times,explant types,initiation and subculture medium formula were compared,and suitable conditions were selected.The results showed that the explants should be selected from tender stems of A.retrofractus,and sterilized with 0.1%mercuric chloride for 8-<10 minutes.The initiation medium formula was MS+6-BA 0.5 mg/L+IBA 1.0 mg/L+sucrose 30 g/L+agar 4.5 g/L,and the induction rate of axillary buds could reach 60%.The optimum subculture medium formula was MS+6-BA 0.6 mg/L+IBA 1.0 mg/L+sucrose 30 g/L+agar 4.5 g/L.The buds induced on the initiation medium could grow normally on this subculture medium,and cluster buds could be formed after 90 days of culture.The establishment of aseptic system of A.retrofractus can provide a foundation for the subsequent development of a complete tissue culture and rapid propagation system of A.retrofractus.
