摘  要:铜是植物体生长发育所必需的一种微量元素,铜缺乏或过量都会对植物的生长不利。杜氏盐藻(Dunaliella salina)是一种富含β-胡萝卜素、甘油等多种营养物质的真核绿藻。本研究分别对f/2培养基(对照组)和5.0 mg/L CuCl2(实验组)中培养72 h的杜氏盐藻进行转录组测序,探讨铜胁迫对盐藻生长发育及不同信号通路的影响。结果获得41418个unigenes,其中注释到NR、COG、KEGG、Swiss-Prot和InterPro数据库的unigene分别有22204、19524、15345、12136和18016个。铜胁迫下共筛查到3799个差异表达基因,其中2350个基因上调,1449个基因下调。对这些差异基因的GO和KEGG富集分析发现,铜胁迫可诱导光合作用、碳固定、铜转运蛋白及热休克蛋白等相关基因的表达,促进杜氏盐藻的生长,增强其对铜的耐受性。杜氏盐藻中发现了多个SNP和SSR结构位点。本研究结果为深入揭示杜氏盐藻铜胁迫下基因差异表达及分子机理提供了平台,也为其后续分子遗传学及表型性状等研究提供一定的理论依据。Copper is a kind of essential trace elements for the growth and development of plants.Green alga Dunaliella salina is rich in beta-carotene,glycerol and other nutrients.In the study,transcriptome sequencing was performed on D.salina cultivated in f/2 culture medium(the control group)and 5.0 mg/L CuCl2(the experimental group)for 72 h,to explore the effects of copper stress on the growth and development of the green alga and different signal pathways.A total of 41418 unigenes were found including 22,204 unigenes annotated in NR,19,524 unigenes in COG,15,345 unigenes in KEGG,12,136 unigenes in Swiss-Prot and 18,016 unigenes in InterPro databases.In addition,3,799 of differentially expressed genes(DEGs)were screened out under copper stress,with up-regulated 2,350 genes and down-regulated 1,449 genes.The GO and KEGG enrichment analysis of these DEGs revealed that copper stress induced the expressions of genes related to photosynthesis,carbon fixation,copper transporter and heat shock proteins,thus promoting the growth of the green alga and enhancing its tolerance to copper.Also,CDS,SNP and SSR structures of these unigene were predicted.The findings will provide a platform for further study the differentially expressed genes and the response mechanism of the green alga to copper stress,even the molecular genetics and phenotype analysis.
