摘 要:为了解决海带配子体人工高密度培养过程中因海水中游离CO_(2)减少及pH升高而导致生长受限的问题,本研究以海带’奔牛’(Sacchatina japonica)雌雄配子体为材料,以不用pH的富营养灭菌海水(10 mg/L N,1 mg/L P)为培养液,通过测量配子体长度,计算相对生长率,比较不同pH对配子体生长的影响,结果显示,pH(P<0.001)、性别(P=0.016<0.05)对海带’奔牛’配子体生长有显著影响,长期处在pH<7培养条件下的雌雄配子体相对生长率显著低于其他2组,雌配子体的相对生长率(4.61%)显著高于雄配子体(3.35%)。初始pH为7.0的处理组与对照组相比,雌雄配子体的相对生长率均无显著差异(P>0.05)。在此基础上,采用在通空气悬浮培养同时补充CO_(2)的方式进行配子体扩增培养,利用纯CO_(2)压力罐(出气量120 L/h),15 L培养液每2 d补充CO_(2)1~2 min,培养液pH范围即可控制在7.0~8.4,以此排除pH对配子体生长不利影响,并避免频繁测定pH对配子体可能造成的混种或杂藻等污染。结果显示,该培养方式下,雌雄配子体的特定生长率(♀15.42%,♂13.70%)显著高于其他2种培养方式。静置培养组雌雄配子体特定生长率(♀10.02%,♂8.12%)最低,其次为通空气悬浮培养组。在3种培养方式下,雌配子体的特定生长率显著高于雄配子体。海带’奔牛’雌雄配子体在3种培养方式下继代培养60 d后,将雌雄配子体按质量比2∶1混合后放于适宜条件下进行发育实验。结果显示,3种扩增培养模式下高密度培养的配子体均可在适宜的条件下正常发育,且发育率无显著差异(P>0.05)。以上研究结果可为利用补充CO_(2)进行海带配子体高密度扩增培养提供理论依据。In order to solve the problem of growth restriction caused by free CO_(2) and pH in seawater during the artificial high-density culture of Saccharina japonica gametophytes,the following experiments were carried out with the female and male gametophytes of S.japonica.The effects of different pH on the growth of male and female gametophytes of S.japonica’Benniu’were studied by measuring the length of gametophytes cultured in pH-free eutrophic sterilized seawater(10 mg/L N,1 mg/L P).The results show that,the effects of pH(P<0.001)and sexuality(P=0.016<0.05)on the growth of gametophytes were very significant.The relative growth rate of female and male gametophytes when the initial pH was 6.0 was significantly lower than that of the other two groups,and the relative growth rate of female gametophytes(4.61%)was significantly higher than that of male gametophyte(3.35%).The relative growth rate of female and male gametophytes in the treatment group with initial pH of 7.0 was not significantly different from that in the control group(P>0.05).On this basis,the method of aerate suspension culture and CO_(2) supply was used to culture gametophytes.Using pure CO_(2) pressure tank(120 L/h)and 15 L culture solution with CO_(2)1~2 min for 2 days,the pH range of the culture solution could be controlled at 7.0~8.4,the adverse effects of pH on the growth of gametophytes were eliminated,and the possible mixture or pollution of gametophytes caused by frequent determination of pH were avoided.The results showed that the specific growth rate of female and male gametophytes(♀15.42%,♂13.70%)was significantly higher than that of the other two methods.The specific growth rate of female and male gametophytes(♀10.02%,♂8.12%)was the lowest in the static culture group,and the next was in the aerate suspension culture group.The development of gametophytes of S.japonica‘Benniu’was studied after 60 days of culture in three different amplification culture conditions.The results showed that the gametophytes cultured in three differe
