摘  要:以湖南省的7个野生云芝菌株为试验材料,基于ITS序列进行分子鉴定以确定样品的种属关系,以MEGA 11.0软件中的邻接法构建系统发育树,采用ISSR分子标记技术对样品进行遗传多样性分析并构建DNA指纹图谱,同时辅以拮抗及栽培试验对结果进行验证。结果表明,7个野生菌株与云芝的同源性为95%~100%;构建的系统发育树显示7个菌株均与云芝聚为1支,且菌株NX4和NX6高度同源。筛选到6条重复性好且条带清晰的ISSR引物,平均每条引物扩增出11条谱带,多态位点百分率为95.5%;ISSR遗传聚类图谱显示在相似性为67%时,7个云芝菌株可聚为2类,其中NX4、NX6归于一类,其他5个菌株归于一类且NX3和NX18遗传差异较小。结合拮抗和栽培试验结果,基于子实体半径尺寸可将NX4和NX6归于一类。Seven wild strains of Trametes versicolor from Hu'nan Province were used as test materials,and the molecular identification was based on ITS sequences to determine the species relationship of the samples,and the phylogenetic tree was constructed by the neighbor joining method in MEGA 11.0 software,and the genetic diversity of the samples was analyzed by ISSR molecular marker technology and DNA fingerprinting was constructed,and the results were verified by antagonism and cultivation tests.The results showed that the homology between the seven wild strains and T.versicdor was 95%-100%;the constructed phylogenetic tree showed that all seven strains clustered with T.versicdor as one branch,and strains NX4 and NX6 were highly homologous.Six ISSR primers with good reproducibility and clear bands were selected,and each primer amplified 11 bands on average,with 95.5%polymorphic sites;ISSR genetic clustering map showed that at 67%similarity,the seven strains of T.versicdor could be clustered into two categories,among which NX4 and NX6 were grouped together,and the other five strains were grouped together and the genetic differences between NX3 and NX18 were small.The results of the antagonism and cultivation tests showed that NX4 and NX6 could be grouped together based on the radius of the substrates.
