摘  要:为研究2021—2022年猪繁殖与呼吸综合征病毒(PRRSV)在山东地区的流行情况,从疑似发病的猪场采集了103份临床样本,采用RT-PCR方法对样品进行检测,并对阳性样品进行GP5和NSP2基因的序列测定、遗传进化分析以及同源性分析。结果显示:103份样品中检测到21份阳性样品,阳性率为20.38%。最终有5个样品能够同时扩增出GP5和NSP2基因序列,遗传进化分析显示,这5个样品中有2个属于谱系(lineage)1;2个属于lineage 5;1个属于lineage 8。各样品之间GP5基因的核苷酸同源性为81.5%~99.3%,NSP2基因的核苷酸同源性为56.9%~99.4%。本研究表明2021—2022年山东地区的PRRSV传播呈现多样性,为PRRSV毒株的遗传变异、演化分析和防控提供了理论基础。To study the prevalence of porcine reproductive and respiratory syndrome virus(PRRSV)in Shandong Province from 2021 to 2022,103 clinical samples were collected of the suspected disease afflicting pigs in different farms.RT-PCR was used to detect the presence of PRRSV in the samples.The results showed that 21 samples were positive,at a positive rate of 20.38%.Then,the GP5 and NSP2 genes of 5 PRRSV strains were amplified and sequenced at the same time.The genetic evolution analysis showed that two strains belonged to lineage 1,another two strains belonged to lineage 5 and one strain belonged to lineage 8.The nucleotide homology of the GP5 and NSP2 genes of the 5 PRRSV strains were 81.5%-99.3% and 56.9%-99.4%,respectively.Our study showed that PRRSV transmission in Shandong Province from 2021 to 2022 was of diversity,which provided a theoretical basis for the genetic variation,evolutionary analysis,and prevention and control of PRRSV.
