摘　　要：旨在调查上海地区奶牛乳房炎大肠杆菌的感染情况及其耐药性和携带的毒力基因,为临床大肠杆菌引起奶牛乳房炎的防控供理论依据。采集上海地区2个牛场60头乳房炎患牛的乳样60份,进行细菌分离培养与鉴定及药敏试验,并检测志贺毒素基因stx1、stx2和紧密黏附素基因eaeA。根据测序结果及BLAST比对确定从60份乳样中共检测出20株大肠杆菌,对氧氟沙星和左氟沙星高度敏感(100%),对氨苄西林的耐药率最高(70%),其次为头孢唑啉(55%)。对其毒力基因的检测发现,这20株大肠杆菌eaeA检出率为100%,没有检测到stx1、stx2毒力基因。研究结果为上海地区部分奶牛场大肠杆菌性乳房炎预防及临床用药提供了理论参考。The infection,drug sensitivity and virulence genes of Escherichia coli in clinical mastitis of dairy cows was investigated in order to provide a theoretical basis for clinical prevention and treatment colibacillosis of mastitis in the Shanghai area.60 milk samples were collected from 60 cows inflicted with mastitis in two dairy cattle farms in Shanghai.Escherichia coli was isolated from the samples and identified by bacteriology,morphology,KOH test,drug sensitivity test and gene sequencing.The virulence factors of stx1,stx2,eaeA were detected by PCR.Twenty Escherichia coli strains were identified by gene sequencing and BLAST analysis.Antibiotic susceptibility determination showed that a total of 20 Escherichia coli strains were identified from the 60 milk samples,which were all 100% highly sensitive to ofloxacin and levofloxacin,and 70%(14/20)of the Escherichia coli strains were highly resistant to ampicillin,and 55%(11/20)were resistance to cefazolin.All of the 20 Escherichia coli strains carried the eaeA gene amplified by PCR.No stx1 or stx2 virulence gene of Escherichia coli was detected by PCR.The present study provided reference for clinical drug treatment and prevention of mastitis in cows in some dairy farms in the Shanghai area.
