摘  要:目的:探究小青龙汤及其方元对寒饮蕴肺证模型大鼠气道黏液高分泌及炎性因子的干预作用。方法:SPF级雄性大鼠56只,通过“形寒+饮冷+冷水浴”法建立寒饮蕴肺证大鼠病理模型。模型建立成功后,随机分为对照组、模型组、小青龙汤组、桂枝甘草组、芍药甘草组、姜辛味组及半夏麻黄组,每组8只,并给予相应药物进行干预。肺功能分析系统测定大鼠FEF50、FVC、MMEF、tI、tE等参数;HE、AB-PAS染色观察大鼠肺组织病理形态学改变;免疫组织化学法检测大鼠肺组织中MUC5AC、AQP5、EGFR、FOXA2的表达;ELISA法检测肺组织中TNF-α、Eotaxin、IL-12、IL-13的含量。结果:(1)模型组大鼠肺内炎性细胞聚集,周围组织充血,肺泡腔内充满水肿液,伴有气道黏液过度分泌,小青龙汤及其方元各组均能不同程度地改善模型大鼠肺组织的病理改变;(2)与对照组相比,模型组大鼠FEF50、FVC、MMEF功能表现下降(P<0.01),tI与tE时间延长,差异具有统计学意义(P<0.01),小青龙汤及其方元各组均能不同程度地改善模型大鼠的肺功能;(3)与对照组相比,模型组大鼠肺组织AQP5(P<0.05)、FOXA2(P>0.05)表达减低,小青龙汤及其方元各组均能在数值上上调其阳性表达;模型组MUC5AC(P<0.05)、EGFR(P>0.05)蛋白表达上调,小青龙汤及其方元各组均能在数值上下调其阳性表达;(4)与对照组相比,模型组大鼠肺组织中TNF-α(P<0.01)、Eotaxin(P<0.05)、IL-13(P<0.01)含量增加,小青龙汤及其方元各组均能不同程度下调其表达;模型组IL-12含量减低(P<0.01),小青龙汤及其方元各组均能不同程度上调其含表达,差异具有统计学意义(P<0.05,P<0.01)。结论:小青龙汤对肺组织MUC5AC、EGFR表达有抑制作用,同时可以降低炎性因子TNF-α、Eotaxin及IL-13分泌,能够上调AQP5、FOXA2的表达,并逆转IL-12的含量减少,从而改善气道黏液高分泌状态、降低气道炎症表现,且小青龙汤所包含的4组方元Objective:To explore the effect of Xiaoqinglong Decoction and its formula components on airway mucus hypersecretion and inflammatory factors in rats with syndrome of cold-fluid accumulating in Lung.Methods:8 rats were randomly chosen from 56 SPF male rats into the control group,the other rats were established the pathological syndrome model of cold-fluid accumulating in Lung by the method of‘cold body+cold drink+cold bath’.After successful modeling,the model rats were randomly divided into the model group,the Xiaoqinglong Decoction group,the Guizhi Gancao group,the Shaoyao Gancao group,the Jiangxinwei group and the Banxia Mahuang group,with 8 rats in each group.Each group was given corresponding intervention.The pulmonary function parameters of FEF50,FVC,MMEF,tI and tE were detected by lung function analysis system.The pathological morphology was observed by HE staining and AB-PAS staining.The expressions of MUC5AC,AQP5,EGFR and FOXA2 in lung tissues were detected by immunohistochemistry.The contents of TNF-α,Eotaxin,IL-12 and IL-13 in lung tissues were detected by ELISA method.Results:In the model group,there were inflammatory cells gathered in the lung tissues and hyperemia in the surrounding tissues;the alveolar cavity was filled with edema fluid,accompanied by excessive secretion of airway mucus.Xiaoqinglong Decoction and its formula components could improve the pathological changes of lung tissues of the model rats to varying degrees.Compared with those in the control group,the pulmonary function of FEF50,FVC and MMEF were decreased(P<0.01),and tI and tE were prolonged in the model group(P<0.01).Xiaoqinglong Decoction and its formula components could improve the pulmonary function of the model rats to varying degrees.Compared with those in the control group,the expressions of AQP5 and FOXA2 were decreased in the model group(P<0.05,P>0.05),and Xiaoqinglong Decoction and its formula components could up-regulate their positive expressions;the expressions of MUC5AC and EGFR were up-regulated in the model gr
