摘  要:目的:基于网络药理学探讨坤泰胶囊治疗卵巢功能下降的相关物质基础及作用机制。方法:从TCMSP数据库提取熟地、黄连、白芍、黄芩、茯苓、阿胶的化学成分,筛选活性成分及潜在靶点。通过Gene Cards数据库挖掘卵巢储备功能下降的疾病靶点,依据药物靶点及疾病靶点构建有效成分靶点与疾病靶点的韦恩图、化合物-靶点网络调控图、PPI网络图,同时进行GO富集分析及KEGG通路富集分析。结果:坤泰胶囊中共筛选出77个活性化合物,对应卵巢储备功能下降112个靶点。通过分析发现,坤泰胶囊治疗卵巢储备功能下降的核心有效活性成分依次为槲皮素、山奈酚、β谷甾醇、汉黄芩素、豆甾醇、蛇床子素等,其治疗卵巢储备功能下降的核心靶点为AKT1、VEGFA、PTGS2、CASP3、TP53,作用通路涉及雌激素信号通路、VEGF信号通路、糖尿病并发症中的糖化终末产物及其受体信号通路等。结论:通过网络药理学方法揭示了坤泰胶囊可通过多个靶点、多条信号通路对卵巢储备功能下降产生治疗效果,为临床用药提供了理论依据。Objective:Based on network pharmacology,to explore the material basis and mechanism of Kuntai Capsules in treating ovarian function decline.Methods:The chemical constituents of Rehmannia glutinosa,Coptis chinensis,Radix Paeoniae Alba,Scutellaria baicalensis,Poria cocos,and donkey-hide gelatin were extracted from the TCMSP database,and the active ingredients and potential targets were screened.Through the Gene Cards database,the disease targets with decreased ovarian reserve function were mined,and the Venn diagrams,compound-target network regulation diagrams,and PPI network diagrams of active ingredient targets and disease targets were constructed based on drug targets and disease targets.GO enrichment analysis and KEGG pathway enrichment analysis.Results:A total of 77 active compounds were screened out from Kuntai Capsules,corresponding to 112 targets for ovarian reserve function decline.Through the analysis,it was found that the core effective active ingredients of Kuntai Capsules in the treatment of ovarian reserve decline were quercetin,kaempferol,β-sitosterol,wogonin,stigmasterol,osthole,etc.,and the core active ingredients in the treatment of ovarian reserve decline The targets are AKT1,VEGFA,PTGS2,CASP3,and TP53,and the action pathways involve estrogen signaling pathways,VEGF signaling pathways,glycation end products and their receptor signaling pathways in diabetic complications,etc.Conclusion:The network pharmacology method reveals that Kuntai Capsules can produce therapeutic effects on ovarian reserve decline through multiple targets and multiple signaling pathways,which provides a theoretical basis for clinical medication.
