扶正散结方联合顺铂对腹水瘤小鼠免疫调节作用的研究

摘　　要：[目的]探讨扶正散结方联合顺铂对H22腹水瘤小鼠的治疗效果及免疫调节作用。[方法]建立H22腹水瘤模型,将40只小鼠随机分为空白对照组、荷瘤对照组、顺铂组、中药联合顺铂组,每日测量小鼠腹围及腹水量,连续给药2周后,采血并处死小鼠。称取瘤质量,计算抑瘤率,比较各组小鼠各脏器质量,采用酶联免疫吸附(ELISA)法测定小鼠血清中肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)和白细胞介素-2(IL-2)含量。[结果]荷瘤组小鼠体质量及腹围较空白对照组迅速增长,大量腹水生成,胸腺质量减轻。与顺铂组小鼠相比,中药联合顺铂组小鼠体质量、腹围增长缓慢,腹水量减少,胸腺质量增加。顺铂组与中药联合顺铂组较荷瘤对照组抑瘤作用明显,中药联合顺铂组的抑瘤率优于顺铂组(P<0.05)。各组荷瘤小鼠血清中TNF-α、IFN-γ、IL-2含量均较空白对照组低(P<0.05)。与顺铂组比较,中药联合顺铂组IFN-γ、IL-2水平升高(P<0.05)。[结论]扶正散结方能够减少肝癌H22腹水瘤小鼠腹水量,通过上调血清中TNF-α、IFN-γ、IL-2水平来促进肿瘤免疫微环境的正性调控,提高小鼠免疫功能,有效抑制肿瘤生长。[Objective]To investigate the therapeutic effect and immunomodulatory effect of Fuzheng Sanjie Recipe combined with cisplatin on H22 ascites tumor mice.[Methods]The H22 ascites tumor model was established,and 40 mice were randomly divided into a blank group,a tumor-bearing control group,a cisplatin group,and a traditional Chinese medicine(TCM)combined cisplatin group.The abdominal circumference and ascites volume of the mice were measured every day,and after continuous administration for 2 weeks,and blood was collected and the mice were sacrificed.Weigh the tumor,calculate the tumor inhibition rate,compare the weight of each organ of each group of mice,and determine the serum tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)and white blood cells in the serum of mice by ELISA method Interleukin-2(IL-2)content.[Results]The body weight and abdominal circumference of the mice in the tumor-bearing group increased rapidly compared with the blank control group,a large amount of ascites was produced,and the weight of the thymus was reduced.Compared with mice in the cisplatin group,the weight and abdominal circumference of the mice in the Chinese medicine combined with cisplatin group increased slowly,the amount of ascites decreased,and the weight of the thymus increased.Compared with the tumor-bearing control group,the cisplatin group and the TCM combined with cisplatin group had anti-tumor effect,and the tumor inhibition rate of the TCM combined with cisplatin group was significantly better than that of the cisplatin group(P<0.05).The serum levels of TNF-α,IFN-γand IL-2 in tumor-bearing mice in each group were lower than those in the blank control group(P<0.05).Compared with the cisplatin group,the levels of Chinese medicine combined with IFN-γand IL-2 were significantly increased(P<0.05).[Conclusion]Fuzheng Sanjie Recipe can reduce the amount of ascites in mice with hepatoma H22 ascites,through up regulating the levels of TNF-α、IFN-γand IL-2 in serum,we can promote the positive regulation of tumor immune mi
