摘 要:醛类化合物是果汁中一类重要的风味物质。本研究以一种新型羟氨类荧光试剂4-羟氨基丁基-7-甲氧基-香豆素(HAMC)为醛类衍生试剂,发展了一种基于硝酮化反应的柱前衍生方法。该方法实现了13种醛类小分子在C18柱上的基线分离且检测限低至0.2 nmol/L。利用该方法结合高效液相色谱-荧光检测(HPLC-FLD)成功从橙汁样品中检测出糠醛、5-羟甲基糠醛、甲醛、乙醛及庚醛。该方法灵敏度高、准确性好且无需固相萃取等复杂前处理,在复杂基质的醛类物质分析中具有良好的应用前景。Aldehydes are a kind of important flavor compounds in fruit juice.In this study,a hydroxylamine fluorescent reagent 4-hydroxyaminobutyl-7-methoxy coumarin(HAMC)was used to derivatize aldehydes to afford nitrone derivatives with high fluorescence intensities followed by HPLC-FLD analysis.After the optimization of derivatization conditions,a nitrone formation derivatization method was proposed with the merits of room-temperature reaction within 30 min.The method was then validated and applied for the analysis of orange juice samples.Furfural,5-hydroxymethylfurfural,formaldehyde,acetaldehyde and heptanaldehyde were successfully detected and quantified with high accuracies.The method has high sensitivity,good accuracy and simple sample pretreatment,providing an efficient approach for the analysis of aldehydes in complex matrix.
