摘 要:建立了1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化-高效液相色谱法(HPLC)同时测定厚朴叶多糖中的6种单糖含量的方法。采用Phenomenex(4.6 mm×250 mm,4μm)为色谱柱,流动相为0.2%磷酸溶液-乙腈(体积比为82∶18),流速为1.0 mL/min,检测波长为245 nm,柱温为35℃。结果显示,基于HPLC法所建立的单糖检测方法,6种单糖成分在各自线性范围内线性关系良好(R^(2)≥0.9994),平均加标回收率为97.17%~100.22%,相对标准偏差(RSD,n=6)为1.67%~2.82%。16批不同产地厚朴叶单糖组成相似,主要是由葡萄糖、甘露糖、半乳糖、鼠李糖和阿拉伯糖组成。主成分分析结果显示,湖北和重庆样品间的单糖组成得到明显区分。该试验所建立的方法操作简便,重复性和准确度高,适用于厚朴叶多糖中单糖成分的含量测定。A method for the simultaneous determination of six monosaccharides in Magnolia officinalis leaf polysaccharide by 1-phenyl-3-methyl-5-pyrazolone(PMP)pre-column derivatization-high performance liquid chromatography(HPLC)was established.The chromatographic column was Phenomenex(4.6 mm×250 mm,4μm).The mobile phase was 0.2%phosphoric acid solution-acetonitrile(82∶18,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 245 nm and the column temperature was 35℃.Based on the monosaccharide detection method established by HPLC,the linear relationship of the six monosaccharide components was good in their linear range(R^(2)≥0.9994),and the average recovery was 97.17%-100.22%(RSD was 1.67%-2.82%,n=6).The monosaccharide composition of Magnolia officinalis leaves from 16 batches of different producing areas was similar,which was mainly composed of glucose,mannose,galactose,rhamnose and arabinose.The results of principal component analysis showed that the monosaccharide composition between Hubei and Chongqing samples was significantly distinguished.The method was simple,reproducible and accurate.It was suitable for the determination of monosaccharide in Magnolia officinalis leaf polysaccharide.
