摘 要:目的建立十味清瘟汤的指纹图谱并进行化学模式识别分析,同时测定其中4种成分的含量。方法利用高效液相色谱法结合《中药指纹图谱相似度评价系统(2012版)》建立10批十味清瘟汤的指纹图谱并进行相似度评价。采用SPSS 26.0软件对共有峰进行系统聚类分析、主成分分析。采用高效液相色谱法测定10批样品中升麻素苷、毛蕊异黄酮、槲皮素、5-O-甲基维斯阿米醇苷的含量。结果在“十味清瘟汤”特征指纹图谱中,共标定11个共有峰,通过与对照品比较指认其中峰4为5-O-甲基维斯阿米醇苷,峰7为毛蕊异黄酮,峰9为升麻素苷,峰10为槲皮素。各批次的相似度均>0.9,聚类分析结果显示10批十味清瘟汤可分为3类,主成分分析表明,在第3个成分累积方差值占92.4%,5种成分在各自范围内线性良好,精密度、重复性、稳定性的RSD值均<2%,加样回收率平均值为98%~101%,RSD为1.2%~2.9%。结论所建立的十味清瘟汤HPLC指纹测定图谱和含量测定方法符合相关规定,结合化学模式分析识别分析可用于十味清瘟汤的质量控制。Objective To establish the fingerprint of Shiwei Qingwen Decoction and analyze its chemical pattern recognition,and determine the contents of its four components.Methods The fingerprints of 10 batches of Shiwei Qingwen Decoction were established by high performance liquid chromatography(HPLC)combined with The Similarity Evaluation System of Traditional Chinese Medicine Fingerprints(2012 Edition)and evaluated.The common peaks were analyzed by SPSS 26.0.The contents of cimicin glycoside,pistil isoflavone,quercetin and 5-O-methylvisamidol glycoside in 10 batches of samples were determined by HPLC.Results In the characteristic fingerprint of Shiwei Qingwen Decoction,a total of 1l common peaks were demarcated.Compared with the reference substance,Peak 4 was 5-O-methylvisamiside,Peak 7 was pistil isoflavone,Peak 9 was cimicin glycoside,and Peak 10 was quercetin.The similarity of each batch was greater than 0.9.The cluster analysis results showed that the 10 batches of Shiwei Qingwen Decoction could be divided into three categories.The principal component analysis showed that the cumulative variance of the third component accounts for 92.4%,the linearity of the five components was good in their respective ranges,the RSD values of precision,repeatability and stability were less than 2%,the average recovery was 98%~101%,and the RSD was 1.2%~2.9%.Conclusion The established HPLC fingerprint and content determination method of Shiwei Qingwen Decoction comply with the relevant regulations.Combined with chemical pattern analysis and identification analysis,it can be used for the quality control of Shiwei Qingwen Decoction.
