摘  要:以六妹羊肚菌为试材,采用继代培养方法,研究了继代培养次数对菌丝的生长特性和TCC-脱氢酶活性的影响,以期为继代培养次数与菌种退化关系研究提供参考依据。结果表明:不同培养方式下,随着继代次数的增加,菌丝生长速率、生物量、TTC-脱氢酶活性均呈下降趋势,要保持较高酶活性和菌丝活力,继代次数应在5次以内,菌丝萌发快、粗壮、前端辐射状,TCC-脱氢酶活性高,且不同菌种培养方式对TTC-脱氢酶活性影响显著,要充分考虑种性特征与生产方式关系。Taking Morchella sextelata as the test material,the effects of mycelial characteristics and TTC-dehydrogenase activity were investigated by repeated subculture,in order to provide reference for the relationship between repeated subculture and strain degradation.The results showed that under different cultivation conditions,the growth rate of mycelial,biomass and TTC-dehydrogenase activity decreased with repeated subculture increased.In order to maintain high enzyme activity and mycelial activity,repeated subculture in five times,and that mycelial germination was fast,thick and radiation in front.In addition,TTC-dehydrogenase activity was strongly and significantly affected by different methods of strain culture,and the relationship between species characteristics and mode of production should be fully considered.
