摘 要:目的探讨桃红四物汤对维甲酸致骨质疏松模型的干预作用。方法2021年11月至2022年2月采用随机数字表法40只SD大鼠分为模型组、空白对照组、桃红四物汤高剂量、低剂量组(0.4、0.2 g/kg)以及阳性药仙灵骨葆胶囊组(0.4 g/kg)。各组连续灌胃30 d。大鼠皮下注射四环素、钙黄绿素标记骨组织。微计算机断层扫描技术分析骨形态计量学参数的变化;苏木素-伊红和番红固绿染色观察股骨病理;抗酒石酸酸性磷酸酶染色检测破骨细胞表达水平;免疫组化法检测股骨骨形态发生蛋白BMP-2表达。结果与模型组比较,桃红四物汤高剂量组大鼠能够改善骨小梁网状结构;骨密度[(0.07±0.02)g·HA^(-1)·cm^(-3)比(0.18±0.04)g·HA^(-1)·cm^(-3)]、骨小梁与骨组织面积比(4.39±0.87比9.02±1.56)、骨小梁宽度[(0.09±0.01)mm比(0.11±0.02)mm]和骨小梁数量[(0.46±0.07)个/毫米比(1.34±0.12)个/毫米]显著升高(P<0.05),骨小梁分离度[(1.27±0.28)mm比(0.86±0.09)mm]显著降低(P<0.05),荧光周长[(1.67±0.30)mm比(3.55±0.82)mm]、骨矿化表面积比(10.28±2.35比14.08±3.74)、双荧光间平均距[(6.93±2.09)µm比(8.76±1.14)µm]、骨矿化沉积率[(0.69±0.21)µm/d比(0.88±0.11)µm/d]增加(P<0.05)、骨形成率[(33.84±25.27)µm^(3)·µm^(-2)·a^(-1)比(45.06±14.05)µm3·µm^(-2)·a^(-1)]增加;破骨细胞数量减少,BMP-2表达增多。结论桃红四物汤能够提高骨质疏松大鼠的骨密度,促进骨组织的动态形成,改善骨微观结构,同时抑制破骨细胞功能。Objective To study the intervention effect of Taohong Siwu decoction on the osteoporosis model of retinoic acid.Methods From November 2021 to February 2022,forty SD rats were randomly assigned into control group,model group,Taohong Siwu decoction low and high dose(0.2,0.4 g/kg)group,and positive drug Xianling Gubao capsule(0.4 g/kg)group according to random number table method,each group was gavaged for 30 days.Subcutaneous injection of tetracycline and calcein labeed bone tissue in rats.The morphometric parameters of femur bone were analyzed by microcomputed tomography;the pathological changes of femur bone were observed by hematoxylin-eosin and saffron solid green staining;the number of osteoclasts was detected by anti-tartrate acid phosphatase staining;the expression of bone morphogenetic protein 2(BMP-2)in femur bone was detected by immunohistochemistry.Results Compared with the model group,the rats in the Taohong Siwu decoctionhigh-dose group were able to improve bone trabecular network structure;bone density[(0.07±0.02)g·HA^(-1)·cm^(-3) vs.(0.18±0.04)g·HA^(-1)·cm^(-3)],bone trabecular to bone tissue area ratio(4.39±0.87 vs.9.02±1.56),bone trabecular width[(0.09±0.01)mm vs.(0.11±0.02)mm]and number of trabeculae[(0.46±0.07)individuals/mm vs.(1.34±0.12)individuals/mm]were significantly higher(P<0.05);trabecular separation[(1.27±0.28)mm vs.(0.86±0.09)mm]was significantly lower(P<0.05),and fluorescence circumference[(1.67±0.30)mm vs.(3.55±0.82)mm],bone mineralization surface area ratio[(10.28±2.35)vs.(14.08±3.74)],mean distance between double fluorescence[(6.93±2.09)µm vs.(8.76±1.14)µm],bone mineralization deposition rate[(0.69±0.21)µm/d vs.(0.88±0.11)µm/d]increased(P<0.05),and bone formation rate[(33.84±25.27)µm3·µm^(-2)·a^(-1) vs.(45.06±14.05)µm^(3)·µm^(-2)·a^(-1)]increased;osteoclast number was reduced and BMP-2 expression was increased.Conclusion Taohong Siwu decoction can increase bone density,promote the dynamic formation of bone tissue,improve bone microstructure,and in
