基于超高效液相色谱-四极杆飞行时间高分辨质谱联用技术的益心舒胶囊化学成分分析与鉴定

摘　　要：目的采用超高效液相色谱-四极杆飞行时间高分辨质谱(UPLC-Q-TOF-MS)联用技术对中药复方益心舒胶囊的化学成分进行分析与鉴定。方法自2021年9―12月,采用Waters ACQUITY UPLC HSS T3(2.1 mm×100 mm,1.8µm)分析益心舒胶囊50%甲醇提取物;流动相0.1%甲酸水-乙腈,梯度洗脱;体积流量0.3 mL/min;柱温30℃。采用电喷雾离子源,在正、负离子模式下采集数据。运用MassLynxV4.1软件,根据化合物保留时间、准分子离子峰、二级碎片等质谱信息,结合对照品和相关文献资料,对益心舒胶囊的化学成分进行鉴定。结果从益心舒胶囊中检测并鉴定出86个化合物,其中皂苷35个、酚酸16个、二萜20个、木脂素9个及其他类别成分6个。结论该方法准确稳定,快速阐明益心舒胶囊的化学成分。益心舒胶囊物质基础的阐明为其质量控制、有效成分确定和治疗机制研究提供了有力支撑。Objective To identify the chemical constituents of Yixinshu capsules by ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF/MS).Methods From September to December 2021,the 50%methanol extract of Yixinshu capsules was analyzed on Waters ACQUITY UPLC HSS T3 column(2.1 mm×100 mm,1.8µm)and eluted with 0.1%formic acid and acetonitrile in gradient elution.The flow rate was set at 0.3 mL/min and the column temperature was maintained at 30°C.By applying software MassLynx V4.1,the chemical constituents of Yixinshu capsules were identified according to the retention times,quasi-molecular ions,and MS/MS ions,and referring to the reference standards and literatures.Results In total,86 compounds were identified in Yixinshu capsules,including 35 saponins,16 phenolic acids,20 diterpenes,9 lignans and 6 other compounds.Conclusion The method is accurate and stable,and quickly elucidates the chemical compositions of Yixinshu capsules.The clarification of the material basis of Yixinshu capsules provids strong support for its quality control,determination of active ingredients,and research on its therapeutic mechanism.
