摘  要:目的:研究健脾补肾方对辐射损伤小鼠脾脏免疫功能损伤的治疗效应。方法:从90只小鼠中随机选择18只为正常组,其余72只小鼠采用6.0 Gy X线一次性全身照射,造模成功后将小鼠随机分为模型组、健脾补肾方大剂量组(200%浓度)、健脾补肾方小剂量组(100%浓度)、胸腺肽组。药物灌胃处理后第7、14、30天分批次取材(每批次每组6只),小鼠称体重,分离小鼠脾脏,称重并计算脾脏指数,TUNEL检测脾脏组织细胞凋亡情况,扫描电子显微镜观察脾脏淋巴细胞结构改变,ELISA法检测外周血细胞因子肿瘤坏死因子-α(TNF-α)、γ-干扰素(IFN-γ)、白细胞介素-2(IL-2)浓度。结果:辐射后第7、14、30天,与正常组同期比较,模型组小鼠脾脏指数显著降低(P<0.01),脾脏组织细胞凋亡率显著升高(P<0.01),脾脏淋巴细胞结构损伤明显,IFN-γ、IL-2浓度显著下降(P<0.01),TNF-α浓度显著升高(P<0.01)。与模型组同期比较,各药物干预组小鼠脾脏指数显著升高(P<0.01,P<0.05),脾脏组织细胞凋亡率显著下降(P<0.01),脾脏淋巴细胞结构损伤减轻,IFN-γ、IL-2浓度显著升高(P<0.01,P<0.05),TNF-α浓度显著下降(P<0.01),其中健脾补肾方小剂量组作用优于健脾补肾方大剂量组(P<0.05,P<0.01)。结论:健脾补肾方可以通过减轻脾脏结构损伤,抑制脾脏组织细胞凋亡,提高IFN-γ、IL-2浓度,抑制TNF-α的过度分泌,从而改善辐射损伤小鼠脾脏免疫功能损伤。Objective:To study the therapeutic effect of Jianpi Bushen Prescription on spleen immune injury in radiation-damage mice.Methods:Eighteen of the 90 mice were randomly selected into the normal group,and the remaining 72 mice were treated with one-time total body irradiation by 6.0 Gy X-ray.After successful modeling,the mice were randomly divided into model group,high-dose Jianpi Bushen Prescription group(200%concentration),low-dose Jianpi Bushen Prescription group(100%concentration),and thymosin group.At the 7th,14th and 30th day after drug intragastric treatment,the mice were collected in batches(6 mice in each batch and group).The mice were weighed and spleen were separated,the spleen index was calculated by weighing,the apoptosis of spleen tissue cell was measured by TUNEL,the structure changes of spleen lymphocytes was observed by scanning electron microscope,and the secretion levels of cytokines tumor necrosis factorα(TNF-α),gamma-interferon(IFN-γ)and interleukin-2(IL-2)were detected by ELISA.Results:On the 7th,14th and 30th day after radiation,compared with the normal group,the spleen index of mice in model group was significantly decreased(P<0.01),the apoptosis rate of spleen tissue cells was significantly increased(P<0.01),the structural damage of spleen lymphocytes was obvious,the concentrations of IFN-γand IL-2 were decreased(P<0.01),and the concentration of TNF-αwas increased(P<0.01).Compared with model group at the same period,spleen index,spleen cell apoptosis rate,splenic lymphocyte structural damage,IFN-γand IL-2 concentrations were significantly increased in drug intervention groups(P<0.01,P<0.05).The concentration of TNF-αwas significantly decreased(P<0.01).The effect of low-dose Jianpi Bushen Prescription group was better than that of high-dose Jianpi Bushen Prescription group(P<0.01,P<0.05).Conclusion:Jianpi Bushen Prescription can alleviate the damage of spleen structure,inhibit the apoptosis of spleen tissue cells,increase the concentrations of IFN-γand IL-2,and inhibit the excessive s
