摘  要:目的探讨人胎盘间充质干细胞(human placenta derived mesenchymal stem cells,HP-MSCs)对变应性鼻炎(AR)小鼠的治疗效果。方法将32只BALB/c小鼠随机平均分为4组(表示为:致敏/激发/治疗):对照组(PBS/PBS/PBS),对照+HP-MSCs组(PBS/PBS/HP-MSCs),AR+PBS组(OVA/OVA/PBS),AR+HP-MSCs组(OVA/OVA/HP-MSCs)。建立卵清蛋白(OVA)联合氢氧化铝诱导的小鼠AR模型,在造模的第20天尾静脉注射0.2 ml PBS或HP-MSCs。评估各组小鼠AR鼻部症状变化;血涂片瑞氏染色观察外周血中嗜酸性粒细胞比例;酶联免疫吸附试验(ELISA)法测定外周血Ig E、Ig G1水平及鼻黏膜白细胞介素4(IL-4)、IL-5、IL-13水平。结果AR+HP-MSCs治疗组AR组小鼠AR行为症状学评分显著减轻(6.25±0.89vs.3.38±0.74,P<0.001);外周血嗜酸性粒细胞比例明显降低[(1.63±1.03)%vs.(4.13±2.57)%,P<0.05];外周血Ig E、Ig G1水平明显降低[(36.89±2.34)ng/ml vs.(57.23±2.81)ng/ml,(21.35±0.80)ng/ml vs.(27.23±0.55)ng/ml,P均<0.05];鼻黏膜IL-4、IL-5、IL-13水平降低[(229.64±12.23)pg/mlvs.(364.17±18.81)pg/ml;(48.70±3.87)pg/mlvs.(68.36±6.88)pg/ml;(41.27±2.48)pg/ml vs.(60.60±4.75)pg/ml,P均<0.05]。结论静脉注射HP-MSCs可以有效治疗小鼠AR,为进一步治疗人AR提供新思路与理论基础。OBJECTIVE To explore the therapeutic effect of human placenta derived mesenchymal stem cells(HPMSCs)on allergic rhinitis(AR)mice.METHODS Thirty-two BALB/c mice were randomly divided into 4 groups(sensitized/challenged/treated):control group:PBS/PBS/PBS;control+HP-MSCs group:PBS/PBS/HP-MSCs;AR+PBS group:OVA/OVA/PBS;HP-MSCs group:OVA/OVA/HP-MSCs.The mouse AR model induced by ovalbumin(OVA)combined with aluminum hydroxide was established,and 0.2 ml PBS or HP-MSCs was injected into the tail vein on the 20th day of modeling.The changes of nasal symptoms in each group were evaluated.The proportion of eosinophils in peripheral blood was observed by Wright staining,and the levels of IgE and IgG1 in peripheral blood and interleukin-4(IL-4),IL-5 and IL-13 in nasal mucosa were measured by enzyme-linked immunosorbent assay(ELISA).RESULTS The behavioral symptom score of mice in AR+HP-MSCs group was significantly lower than that in AR group(6.25±0.89 vs.3.38±0.74,P<0.001).The proportion of eosinophils in peripheral blood was significantly lower than that in AR group[(1.63±1.03)%vs.(4.13±2.57)%,P<0.05].The levels of IgE and IgG1 in peripheral blood decreased significantly[(36.89±2.34)ng/ml vs.(57.23±2.81)ng/ml,(21.35±0.80)ng/ml vs.(27.23±0.55)ng/ml,P<0.05].The levels of IL-4,IL-5 and IL-13 in nasal mucosa decreased[(229.64±12.23)pg/ml vs.(364.17±18.81)pg/ml;(48.70±3.87)pg/ml vs.(68.36±6.88)pg/ml;(41.27±2.48)pg/ml vs.(60.60±4.75)pg/ml,P<0.05].CONCLUSION Intravenous injection of HPMSCs could effectively treat AR in mouse model,which provides a new idea and theoretical basis for further treatment of human AR.
