摘 要:为探究微囊藻毒素(Microcystin-LR,MC-LR)对凡纳滨对虾(Litopenaeus vannamei)肝胰腺毒性作用机理,通过血窦注射法,检测注射MC-LR后凡纳滨对虾肝胰腺和肌肉中毒素含量,肝胰腺免疫酶活力、细胞凋亡与周期的变化及凋亡相关基因表达量情况。结果表明,注射MC-LR后,肝胰腺与肌肉中毒素含量均在1 h达到最大值,分别为(6.12±0.45)、(5.00±0.19)μg·kg^(-1),且均极显著高于对照组和其他时间点(P<0.01);碱性磷酸酶(AKP),酸性磷酸酶(ACP)和溶菌酶(LZM)活力分别在8、24和8 h达到最高,分别比对照组增加23.8%、27.3%和124%,且均显著高于对照组(P<0.05);细胞凋亡率和G1期细胞所占比例显著上升(P<0.05),S、G2期细胞所占比例下降;CYC、AIF、Caspase-3和p53基因均呈先升后降趋势,CYC和p53相对表达量在24 h达到最高,AIF和Caspase-3相对表达量在48 h达到最高。研究结果表明,MC-LR进入对虾体内后,可快速转运到肝胰腺并影响其主要免疫酶活力,引发应激反应,进而引起细胞凋亡并诱导细胞周期阻滞于G1期,抑制细胞增殖。研究为探明MC-LR对凡纳滨对虾肝胰腺的毒性作用机制以及对虾健康养殖提供数据参考。To investigate the mechanism of Microcystin-LR(MC-LR)toxicity on the hepatopancreas of Litopenaeus vannamei,the toxin content in the hepatopancreas and muscles of L.vannamei,changes in liver pancreatic immune enzyme activity,cell apoptosis and cycle,and expression of apoptosis related genes were detected by sinus injection method.The results showed that after injection of MC-LR,the toxin content in hepatopancreas and muscle reached the maximum at 1 h,which was(6.12±0.45)and(5.00±0.19)μg·kg^(-1),respectively,and was significantly higher than that of the control group and the other time-points(P<0.01);the activities of AKP,ACP and LZM reached their highest levels at 8,24 and 8 h,respectively,with an increase of 23.8%,27.3%and 124%compared to the control group,and were significantly higher than those of the control group(P<0.05);the apoptosis rate and the proportion of G1 phase cells increased significantly(P<0.05),while the proportion of S and G2 phase cells decreased;the CYC,AIF,Caspase-3 and p53 genes showed a trend of first increasing and then decreasing,with relative expression levels reaching their highest at 24,48,48 and 24 h,respectively.The results showed that after MC-LR entered the body of shrimp,it could be quickly transported to hepatopancreas and affect the activity of its main immune enzymes,causing stress reaction,and then causing cell apoptosis,which induced cell cycle arrest in G1 phase,as well as inhibited cell proliferation.This study provided data reference for exploring the toxic mechanism of MCLR on the hepatopancreas of L.vannamei and the healthy growth of shrimp.
