摘 要:为探究大豆食心虫未滞育-滞育-滞育解除过程中虫体内激素的变化,进而揭示大豆食心虫滞育机理,分析了经4,10,15,20和25℃及室外自然温度处理后的大豆食心虫滞育前及不同滞育阶段其体内调节滞育的激素,包括蜕皮激素、保幼激素、促前胸腺激素,以及催化蜕皮激素合成酶-细胞色素P450的表达动态变化。结果表明,自然状态下进入滞育的大豆食心虫体内蜕皮激素、保幼激素、促前胸腺激素、细胞色素P450含量与未滞育阶段相比差异显著(P<0.05),分别下降了23.22μg·mL^(-1)、73.83 pg·mL^(-1)、120.12 pg·mL^(-1)和4.74 ng·mL^(-1);不同温度诱导进入滞育30 d后,10℃处理组虫体内蜕皮激素、保幼激素、促前胸腺激素、细胞色素P450含量均显著高于其他温度处理组,含量分别为253.22μg·mL^(-1)、210.72 pg·mL^(-1)、319.85 pg·mL^(-1)和12.53 ng·mL^(-1)。不同滞育诱导温度处理组在滞育初期(11月)、滞育中期(2月)、滞育后期(6月)虫体内的蜕皮激素、保幼激素、细胞色素P450含量均表现出先下降后升高的趋势,且在2月份均达到最低值,表明2月可能是大豆食心虫由滞育向滞育解除的转折关键期。In order to explore the changes of hormones and hormone-related enzymes in soybean pod borer during the process of non-diapause-diapause-diapause removal,and further reveal the diapause mechanism of soybean pod borer,this experiment analyzed the hormones regulating diapause in soybean pod borer before diapause,after diapause at 4,10,15,20 and 25℃and at different diapause stages.Dynamic changes in the expression of ecdysone,juvenile hormone,prothymotropic hormone,and the catalytic ecdysone synthetase-cytochrome P450.The results showed that,the contents of ecdysone,juvenile hormone,prothymotropic hormone and cytochrome P450 in natural diapause were significantly different from those in non-diapause stage(P<0.05),and decreased by 23.22μg·mL^(-1),73.83 pg·mL^(-1),120.12 pg·mL^(-1),4.74 ng·mL^(-1)respectively;After 30 days of diapause induced by different temperatures,the contents of ecdysone,juvenile hormone,prothymotropic hormone and cytochrome P450 in 10℃group were significantly higher than those in other temperature groups(P<0.05),with the contents of 253.22μg·mL^(-1),210.72 pg·mL^(-1),319.85 pg·mL^(-1),12.53 ng·mL^(-1)respectively.The contents of ecdysone,juvenile hormone and cytochrome P450 in the treatment groups with different diapause induction temperature at the early stage of diapause(November),the middle stage of diapause(February)and the late stage of diapause(June)showed a trend of first decreasing and then increasing.They all reached the lowest value in February,indicating that February may be the key period of transition from diapause to diapause.
