分枝(瓜)列当寄生烟草过程中转移miRNA筛选及其功能预测

摘  要:旨在明确分枝列当(Phelipanche aegyptiaca Pers.)与寄主之间是否存在miRNA转移,同时筛选出转移的miRNA并对其功能进行预测。以分枝列当-普通烟(Nicotiana tabacum Linn.)为研究体系,分别对寄生有分枝列当的烟草根部、寄生接触点和列当茎秆取样进行sRNA测序和分析。结果表明,烟草在被分枝列当寄生后,与对照相比,产生49个差异miRNA,包括17个已知miRNA和32个新预测的miRNA,其中30个miRNA显著下调表达,靶mRNA主要参与植物根系的生长和水杨酸的合成,19个miRNA显著上调表达,靶mRNA主要参与矿质元素吸收和积累及脱落酸的表达。筛选鉴定到3个烟草miRNA转移至分枝列当,靶基因主要参与植物抗逆和过敏性坏死反应;筛选鉴定到2个miRNA由分枝列当转移至烟草,其靶基因功能主要与水杨酸的产生和植物根系生长有关。表明miRNA在分枝列当寄生寄主时存在双向转移,从miRNA角度为分枝列当与烟草互作的分子机制研究提供了候选,也为分枝列当的防治提供了新思路。The purpose of the study is to clarify and screen miRNA transfer between Phelipanche aegyptiaca Pers.and its host,and to predict the miRNAs function.The P.aegyptiaca-Nicotiana benthamiana L.system was used,the parasitized root of tobacco,attachment site and stem of P.aegyptiaca were collected to perform sRNA sequencing and analysis.The results indicated that compared with non-infected tobacco roots,49 differentially expressed miRNAs were screened out after parasitized by P.aegyptiaca,which included 17 known miRNAs and 32 novel miRNAs.In the 49 differentially expressed miRNAs,30 miRNAs were significantly down-regulated expression,and their target mRNAs were mainly involved in plant root growth and salicylic acid synthesis;the expression of other 19 miRNAs was significantly up-regulated,its target mRNAs were mainly involved in mineral element absorption and accumulation and expression of abscisic acid(ABA).We also found that 3 miRNAs transferred from N.benthamiana to P.aegyptiaca,its target genes had close relationship with plant stress resistance and hypersensitive response(HR).Contrarily,2 miRNAs involved in salicylic acid(SA)biosynthesis and plant root growth were transferred from P.aegyptiaca to N.benthamiana.In conclusion,the miRNAs transfer between P.aegyptiaca and the host is bidirectional transfer.The findings in this study provide a novel proof for molecular mechanism of the interaction between P.aegyptiaca and N.benthamiana from the perspective of miRNA,and a new idea for control of P.aegyptiaca.
