摘 要:旨在鉴定小菜蛾Plutella xylostella的Hsp20家族基因,明确其在8℃和4℃低温条件下的表达模式。通过生物信息学手段,从小菜蛾染色体级基因组中鉴定出17条Hsp20家族基因,分布在5条染色体,其中11个PxHsp20s位于27号染色体;开放阅读框(ORF)长度为501~1002,理论等电点5.65~8.05,蛋白分子质量18.80~36.60 ku。将对照组(26℃恒温)、短期低温组(4龄幼虫8℃24 h,以CS表示)、适应后短期低温组(3龄幼虫15℃适应后4龄幼虫8℃24 h,以CA表示)的小菜蛾4龄幼虫转录组进行分析,仅PxHsp20-8差异表达。利用RT-qPCR对温和低温(8℃、4℃)、低温暴露时间(24 h、48 h、96 h)、4龄幼虫前是否经历适应(CS和CA处理)不同组合条件下小菜蛾4龄幼虫的PxHsp20-8基因表达量进行检测。结果表明,处理温度高低和处理时长对PxHsp20-8表达量无显著影响,4龄前经历15℃适应对表达量有显著影响;4龄幼虫CS组8℃48 h、4℃48 h、4℃96 h处理后PxHsp20-8表达量显著高于26℃对照,而CA组8℃24 h、8℃48 h、4℃96 h处理后显著低于对照。相同温度—时间组合下,CS组8℃24 h、8℃48 h、4℃48 h、4℃96 hPxHsp20-8相对表达量显著高于CA组相应低温处理。Aim to identify genes inHsp20families of Plutella xylostella and toclarify the expression pattern of them at 8℃and 4℃.ElevenPxHsp20swere identified from the chromosome-level genome located at 5 chromosomes,eleven of them was located at Chr27.The length of open reading frames of 17Hsp20sranged from 501 to 1002 bp,and its theoretical isoelectric point of deduced proteinswas between 5.65 to 8.05,the molecular mass of deduced proteins was between 18.80 to 36.60 ku.OnlyPxHsp20-8was significantly differentially expressed based on transcriptomic data from the control(constant 26℃),short-time low temperature(4th instar larvae exposed at 8℃for 24 h,represented by CS),and short-time low temperature after adaptation(4th instar larvae exposed at 8℃for 24 h after 3th instar larvae experienced 15℃,represented by CA).RT-qPCR was used to determine thePxHsp20-8expression level of 4th instar larvae of Plutella xylostella under different conditions of mild low temperature(8℃,4℃),cold exposure duration(24 h,48 h,96 h),and whether the 4th instar larvae of Plutella xylostella experienced adaptation or not(CS or CA).The results showed that both the value of low temperature and the duration of low temperature had no significant effects on therelative expression ofPxHsp20-8,and whether the 4th instar larvae of Plutella xylostella experienced adaptation had significant effect on the expression value.The relative expressions ofPxHsp20-8of CS groups with 8℃48 h,4℃48 h,and 4℃96 h were significantly higher than control,while those of CA groups with 8℃24 h,8℃48 h,4℃96 h were significantly lower than control.In the same temperature-duration combination,the relative expression level in CS treatment was significantly higher than in CA treatment with 8℃24 h,8℃48h,4℃48 h,and 4℃96 h.
