摘 要:探究砂生槐子生物碱对LPS诱导的小鼠急性肺炎的保护作用。将48只SPF雄性昆明小鼠随机分为对照组(Control组)、脂多糖组(LPS组)、地塞米松组(DXMS组,0.8 mg/kg)、砂生槐子生物碱低剂量组(SSHD组,0.34 mg/kg)、砂生槐子生物碱中剂量组(SSHZ组,0.68 mg/kg)、砂生槐子生物碱高剂量组(SSHG组,1.36 mg/kg),每组8只,给药7 d后,通过滴鼻LPS(5 mg/kg)建立小鼠急性肺炎模型。结果表明,SSHD组、SSHZ组、SSHG组小鼠WBC、LYM、GRA显著降低(P<0.05),HGB、RBC、PLT显著升高(P<0.05);SSHD组、SSHZ组小鼠脾系数显著降低(P<0.05),SSHD组、SSHZ组、SSHG组小鼠肺系数、胸腺系数显著降低(P<0.05);SSHD组、SSHZ组、SSHG组小鼠血清中TNF-α、IL-6含量显著降低(P<0.05),CRP、IL-1β含量降低;SSHD组、SSHZ组与SSHG组小鼠肺组织中丙二醛(MDA)显著降低(P<0.05),超氧化物歧化酶(SOD)显著升高(P<0.05);各给药组小鼠肺脏的病理损伤均有不同程度改善,其中SSHG组保护效果最好,SSHD组保护效果次之;另外,SSHD组、SSHZ组与SSHG组小鼠肺组织SFTPC染色升高,NF-κB P65与TLR4染色降低。说明砂生槐子生物碱对LPS诱导的小鼠急性肺炎具有较好的保护作用。To explore the protective effect of Sophora moorcroftiana alkaloid on acute pneumonia induced by LPS in mice,forty-eight SPF male Kunming mice were randomly divided into control group,lipopolysaccharide group(LPS group),dexamethasone group(DXMS group,0.8 mg/kg),low-dose Sophora moorcroftiana alkaloid group(SSHD group,0.34 mg/kg),middle-dose Sophora moorcroftiana alkaloid group(SSHZ group,0.68 mg/kg)and high-dose Sophora moorcroftiana alkaloid group(SSHG group,1.36 mg/kg),with 8 mice in each group.After 7 days of administration,LPS(5 mg/kg)was administered through nose.The results showed that WBC,LYM and GRA in SSHD group,SSHZ group and SSHG group decreased significantly(P<0.05),while HGB,RBC and PLT increased significantly(P<0.05).Spleen coefficients of SSHD group and SSHZ group decreased significantly(P<0.05),lung coefficients and thymus coefficients of SSHD group,SSHZ group and SSHG group decreased significantly(P<0.05).The levels of TNF-αand IL-6 in sera of SSHD group,SSHZ group and SSHG group decreased significantly(P<0.05),and the levels of CRP and IL-1βdecreased.MDA in lung tissue of mice in SSHD group,SSHZ group and SSHG group decreased significantly(P<0.05),while SOD increased significantly(P<0.05).The pathological damage of mouse lungs in each treatment group was improved to varying degrees,among which SSHG group had the best protective effect,followed by SSHD group.In addition,SFTPC staining in lung tissue of mice in SSHD group,SSHZ group and SSHG group increased,while NF-κB P65 and TLR4 staining decreased.It showed that Sophora moorcroftiana alkaloid has a good protective effect on LPS-induced acute pneumonia in mice.
