摘 要:目的:研究聚己内酯/钛酸钡[PCL/BaTiO3(PCL/BT)]纳米纤维膜的制备及其力学性能和细胞活性。方法:通过加入BaTiO3,利用静电纺丝技术制成PCL、PCL/3%BT、PCL/6%BT和PCL/9%BT纳米纤维膜。扫描电镜(SEM)观察膜片形貌及间充质干细胞的生长形态,力学性能实验检测材料的机械性能。CCK-8法检测间充质干细胞在膜上7 d间充质干细胞的增殖情况。SEM下观察细胞活力,探究骨髓间充质干细胞在电纺纳米纤维膜上的生长情况。Calcein-AM/PI染色比较间充质干细胞与纳米纤维膜复合培养7 d时细胞存活情况。结果:SEM观察结果显示,纯PCL静电纺丝膜纤维表面光滑,未发现凹陷或凸出,PPCL/3%BT静电纺丝膜纤维表面偶有突出,PCL/6%BT静电纺丝膜纤维变得稀疏,PCL/9%BT静电纺丝膜纤维表面有明显的沉淀物,并且明显可以看出PCL/6%BT的孔隙率优于其他组。间充质干细胞在PCL/6%BT膜上的细胞增殖和活性显著优于其他3种膜片,细胞分布更密集。结论:本研究成功地利用静电纺丝技术制备出PCL、PCL/3%BT、PCL/6%BT和PCL/9%BT纳米纤维膜,该纳米纤维膜具有良好机械性能,并具有促进细胞增殖能力。Objective:To study the preparation,mechanical properties and cellular activities of polycaprolactone/BaTiO3[PCL/BaTiO3(PCL/BT)]nanofiber membranes.Methods:PCL,PCL/3%BT,PCL/6%BT and PCL/9%BT nanofiber membranes were made by adding BaTiO3 using electrospinning technology.Scanning electron microscopy(SEM)observed the diaphragm morphology and the growth morphology of bone marrow mesenchymal stem cells(MSCs),and mechani-cal properties tested the mechanical properties of materials.MSC proliferation of bone marrow on the membrane at 7 days was measured by CCK-8 assay.Cell viability was observed under SEM to explore the growth of bone marrow mesenchymal stem cells(BMMSCs)on electrospun nanofiber membranes.Calcein-AM/PI staining was used to compare cell survival of bone marrow MSCs in compound culture with nanofiber membranes for 7 d.Results:SEM results showed that the surface of pure PCL electrospinning film fiber was smooth with no depression or protrusion,PCL/3%BT electrospinning film fiber surface had protrusion occasionally,PCL/6%BT electrospinning film fiber became sparse,and PCL/9%BT electrospinning film fiber surface was precipitated obviously.It was obvious that the porosity of PCL/6%BT was better than that of other groups.Bone marrow MSCs had significantly better cell proliferation and activity on PCL/6%BT membranes than the other three membranes and a more dense cell distribution.Conclusion:In this study,PCL,PCL/3%BT,PCL/6%BT and PCL/9%BT nanofiber membranes are successfully prepared using electrospinning technology.The nanofiber membranes have good mechanical properties and have the ability to promote cell proliferation.
