摘  要:目的:探讨环状RNA hsa_circ_0001801在先天性心脏病(CHD)相关肺动脉高压(PAH)患儿中的表达及其临床意义。方法:收集30例CHD和30例PAH-CHD患儿全血样本,并收集14例健康儿童全血样本作为对照组,采用RT-qPCR检测hsa_circ_0001801在不同分组中的表达量,并与临床资料进行相关性分析,通过Logistic回归分析确定PAH-CHD的影响因素。结果:PAH-CHD组hsa_circ_0001801相对表达量低于对照组和CHD组(P<0.05),而CHD组与对照组无明显差异。Spearman相关分析显示,hsa_circ_0001801表达与肌酸激酶同工酶呈正相关关系(r=0.393,P=0.002)。多因素Logistic回归分析显示,hsa_circ_0001801为PAH-CHD的保护因素(OR=0.465,95%CI:0.257~0.844,P=0.012)。结论:hsa_circ_0001801在PAH-CHD患儿全血中异常表达,具有作为PAH-CHD生物标志物的潜力。Objective:To explore the expression and clinical significance of circular RNA hsa_circ_0001801 in whole blood of children with pulmonary arterial hypertension(PAH)associated with congenital heart disease(CHD).Methods:The whole blood samples of 30 children with CHD and 30 children with PAH-CHD were collected,and the whole blood samples of 14 healthy children were collected as the control group.The expression levels of hsa_circ_0001801 in different groups were detected by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR),correlation analysis was performed with the clinical data,and the influencing factors for PAH-CHD were determined by logistic regression analysis.Results:The relative expression level of hsa_circ_0001801 in PAH-CHD group was lower than that in control group and CHD group(P<0.05),while there was no significant difference between CHD group and control group.Spearman correlation analysis showed a positive correlation between the expression of hsa_circ_0001801 and creatine kinase-MB(CK-MB)(r=0.393,P=0.002);multivariate logistic regression analysis showed hsa_circ_0001801 was a protective factor for PAH-CHD(OR=0.465,95%CI:0.257-0.844,P=0.012).Conclusion:hsa_circ_0001801 is aberrantly expressed in whole blood of children with PAH-CHD and has potential as a biomarker for PAH-CHD.
