摘  要:目的探究Shh信号通路的异常激活对肾脏纤维化的影响。方法用10月龄快速老化(SAMP)8小鼠与同龄抗快速老化(SAMR)1小鼠进行比较,采用苏木素-伊红(HE)、马松(Masson)染色方法观察肾小球、肾小管间质病理形态改变和纤维化程度;免疫荧光法检测Shh、平滑蛋白(Smo)、胶质瘤相关癌基因同源物(Gli1)蛋白的表达。结果与SAMR1小鼠相比,SAMP8小鼠肾脏可见肾小球及小管间质纤维化明显,Shh、Smo、Gli1蛋白表达显著上调(P<0.05)。结论衰老肾脏纤维化的程度可能与Shh信号通路的异常活化密切相关。Objective To investigate the effect of abnormal activation of Shh signaling pathways on renal fibrosis.Methods Rapid aging(SAMP)8 mice at 10 months of age were compared with anti-rapid aging(SAMR)1 mice at the same age,hematoxylin-eosin(HE)and Masson staining were used to observe the pathological structural changes and degree of fibrosis of renal glomerular and tubulointerstitial.The expressions of Shh,smoothing protein(Smo)and glioma-associated oncogene homolog(Gli)1 protein were detected by immunofluorescence method.Results Compared with SAMR1 mice,SAMP8 mice exhibited a more significant fibrosis of glomerulus and tubulointerstitium,and the expressions of Shh,Smo and Gli1 protein were significantly increased(P<0.05).Conclusions The degree of fibrosis in the aging kidney might be closely related to the abnormal activation of the Shh signaling pathway.
