摘  要:本研究从百合小鳞茎发育不同阶段蛋白质组数据中,筛选出调控鳞茎发育的上调表达基因LdGASA1,对其进行克隆、生物信息学分析及表达分析。序列分析显示LdGASA1基因全长725 bp,编码区CDS序列为336 bp,编码111个氨基酸。结构域分析显示,LdGASA1第52至第111个氨基酸含有一个典型的GASA保守结构域。其理化性质分析显示,编码蛋白质理论等电点(pI)为9.1,不稳定系数为35.14,属于稳定的疏水性蛋白,亚细胞定位预测该蛋白最可能定位在细胞质中,其二级结构主要为无规则卷曲(65.86%)和α-螺旋(22.52%),另外含有少量延伸链(12.60)和β-折叠(9.01%)。通过RT-qPCR分析发现LdGASA1基因在小鳞茎出现前表达量相对较低,主要在小鳞茎形态建成和发育阶段表达,且在小鳞茎形态基本建成(35 d)时表达量达到最高。本研究结果为探索GASA基因调控百合生长发育的机制打下基础。In this study,the upregulated LdGASA1 gene regulating bulb development was screened from the proteomic data at different stages of lily bulb development,and its cloning,bioinformatics analysis and expression analysis were carried out.Sequence analysis showed that the full length of LdGASA1 gene is 725 bp,and the CDS sequence is 336 bp,encoding 111 amino acids.Domain analysis showed that the 52~111 amino acids of LdGASA1 contained a typical GASA conserved domain.It is a stable hydrophobic protein with a theoretical isoelectric point(pI)of 9.1 and an instability coefficient of 35.14.It is located in the cytoplasm,and its secondary structure is dominated by irregular coils(65.86%)and α-helix(22.52%),with a small amount of extended chains(12.60)and β-folded(9.01%).RT-qPCR analysis showed that LdGASA1 gene was not expressed before the appearance of bulblets,but was mainly expressed at the morphogenesis and development stages of bulblets,and the expression level reached the highest at the basic morphogenesis of bulblets(35 d).The results of this study laid a foundation for exploring the mechanism of GASA gene regulating the growth and development of lily.
