长茎葡萄蕨藻多糖的理化性质及其对免疫抑制小鼠的调节作用

摘  要:【目的】为促进长茎葡萄蕨藻(Caulerpalentillifera)的开发利用,研究长茎葡萄蕨藻多糖的理化性质和对免疫抑制小鼠的调节作用。【方法】以海南养殖的长茎葡萄蕨藻为研究对象,采用水提醇沉和离子交换柱层析获得多糖成分,用高效凝胶渗透色谱和高效液相色谱法分析多糖分子质量和单糖组成等理化性质,并从动物水平研究多糖对免疫抑制小鼠的免疫调节和抗氧化活性。【结果与结论】长茎葡萄蕨藻粗多糖产率为5.04%,粗多糖脱蛋白后,经强阴离子交换柱层析纯化得到主要多糖组分CP0。理化性质分析表明,CP0的总糖质量分数为70%,硫酸基质量分数为11%;主要由甘露糖、半乳糖和木糖组成,比例为4.4∶4.0∶1.4,还含有微量的葡萄糖。表明CP0是硫酸化的木糖半乳甘露聚糖,相对分子质量为471000。原子力显微镜显示,CP0为直径约100 nm的近球体结构。CP0对免疫抑制小鼠脾脏指数、胸腺指数有显著影响(P<0.05),对小鼠白介素(IL2、IL4、IL10)、干扰素(IFN)、肿瘤坏死因子(TNF)、肠道分泌型免疫球蛋白A(SIgA)等6种免疫因子有显著调节作用(P<0.05),对总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、丙二醛(MDA)活性等抗氧化指标有显著影响(P<0.05);通过提高小鼠肠道SIgA含量,发挥肠道免疫调节作用,对环磷酰胺引起的免疫抑制有较好的修复作用,可通过提高免疫抑制小鼠的脾脏指数和胸腺指数恢复免疫平衡,并通过增加体内抗氧化酶的活性提高机体抗氧化水平的能力。【Objective】This study aimed to explore the immnoregulation activity of polysaccharides from Caulerpa lentillifera,so as to make better use of C.lentillifera.【Method】Polysaccharides were extracted from cultured C.lentillifera in Hainan Province by water extraction,alcohol precipitationand ion exchange column chromatography..The physicochemical properties of the polysaccharides were analyzed by high performance gel permeation chromatography and high performance liquid chromatography.The immune regulation and antioxidant activity of polysaccharides on immunosuppressed mice were studied.【Result and Conclusion】The yields of the polysaccharides were 5.04%.The main polysaccharide fraction CP0 was purified by Q Sepharose Fast Flow ion-exchange.The physicochemical analysis showed that the total sugar content and sulfate content of the CP0 was 70%and 11%,respectively.The monosaccharide composition showed that CP0 was mainly composed of mannose,galactose and xylose at the ratio of 4.4∶4.0∶1.4.It indicated that CP0 was a sulfated xylogalactomannan.The average relative molecular weight of CP0 by gel permeation chromatography was estimated to be about 471000.Atomic force microscopy(AFM)shows that the morphology of CP0 is a near-spherical structure with a diameter of about 100 nm.The immunoregulatory activity of CP0 showed it had good immunoregulation effect:significant effects on spleen index and thymus index of immunosuppressed mice(P<0.05).It could significantly regulate 6 immune factors including interleukin(IL2,IL4,IL10),interferon(IFN),tumor necrosis factor(TNF)and intestinal secreted immunoglobulin A(SIgA)in mice(P<0.05).It had significant effects on antioxidant indexes such as total antioxidant capacity(TAOC),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT),and malondialdehyde(MDA)activity(P<0.05).These results indicated that CP0 played a good role in intestinal immune regulation by increasing the content of intestinal SIgA in mice,had a good repair effect on the immunosuppression c
