摘  要:【目的】探究骨形态发生蛋白7(BMP7)在香港牡蛎(Crassostrea hongkongensis)生长发育过程中的作用。【方法】通过聚合酶链式反应(PCR)获得香港牡蛎ChBMP7基因核心编码序列(CDS),并对其进行生物信息学分析;采用实时荧光定量PCR(qRT-PCR)技术检测ChBMP7基因在香港牡蛎不同组织和不同发育阶段中的表达变化。【结果】香港牡蛎ChBMP7基因CDS序列长度为1254 bp,共编码417个氨基酸,具有TGF-β超家族结构域(316Cys~417His),与太平洋牡蛎(Crassostrea gigas)BMP7氨基酸序列同源性(98.56%)最高。ChBMP7基因在香港牡蛎8种组织中均有表达,其中外套膜组织中ChBMP7基因相对表达量最高,其次分别为闭壳肌、唇瓣、性腺、消化腺、鳃、血窦、肠。ChBMP7基因在香港牡蛎不同生长发育时期均有表达,ChBMP7基因在卵裂期和担轮幼虫时期的相对表达量显著高于其他发育时期(P<0.05)。【结论】ChBMP7基因参与了香港牡蛎早期生长发育和外套膜相关分泌调控过程。【Objective】To investigate the role of bone morphogenetic protein 7(BMP7)in the growth and development of the Crassostrea hongkongensis.【Method】The ChBMP7 coding sequence(CDS)was cloned by polymerase chain reaction in C.hongkongensis,and was analyzed by bioinformatics.The expression of ChBMP7 in different tissues and developmental stages of C.hongkongensis was detected by real-time fluorescence quantitative PCR.【Result】The sequence of ChBMP7 CDS was 1254 bp,which encoded 417 amino acids.It included TGF-βsuperfamily domain(316Cys-417His).ChBMP7 was highly homologous(98.56%)with that of Crassostrea gigas.ChBMP7 gene was expressed in 8 tissues,and the relative expression was higher in labial palps,followed by the closed shell muscle,labellum,gonad,digestive gland,gill,blood sinus and intestine,respectively.ChBMP7 gene was expressed in all developmental stages,and the relative expression in cleavage and trochophore larvae stages was significantly higher than that in other stages(P<0.05).【Conclusion】ChBMP7 gene is involved in the regulation of the early growth and mantle related secretion of C.hongkongensis.
