正常高值血压痰湿壅盛证动物模型建立与评价

摘　　要：目的:建立正常高值血压痰湿壅盛证大鼠模型及其评价方法。方法:20只WKY雄性大鼠随机分为空白组和模型组各10只。空白组予蒸馏水灌胃,普通饲料喂养;模型组予1 mL/100 g的6%盐水灌胃和高脂饲料饲养,建立正常高值血压痰湿壅盛证模型,并测量21个一般状况评价指标(大鼠应激度、毛发润泽度、大便次数等)、16个体表特征指标(收缩压、舒张压、心率等)、4个血清生化指标(甘油三酯、总胆固醇、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇)和15个行为学指标数据(大鼠旷场实验总路程、高架十字迷宫总路程等),运用正交偏最小二乘判别分析法(OPLS-DA)评价模型。结果:第8周,大鼠血压和宏观体表特征指标达标。第20周,行为学指标出现显著性差异。通过模型变量重要性投影判别筛选出一般状况评价总分、10个体表特征指标(收缩压、舒张压、平均动脉压、心率、体质量、进食量、饮水量、大便含水量、面温、爪色r值)和7个行为学指标(痛阈值、旷场实验:总路程和外周路程、十字迷宫实验:总路程、进入中心次数、封闭臂路程和进入封闭臂次数)的数值大于1。结论:通过高脂高盐饲料喂养20周的WKY大鼠可作为符合正常高值血压痰湿壅盛证的动物模型,OPLS-DA适用于该模型的评价。Objective:To establish the rat model of high-normal blood pressure with excessive phlegm-dampness syndrome and its evaluation method.Methods:A total of 20 WKY male rats were randomly divided into blank group and model group,with 10 rats in each group.The blank group was given distilled water and ordinary feed.The model group was given 1 mL/100 g 6%saline and fed with high fat diet.To establish the animal model of high-normal blood pressure with excessive phlegm-dampness syndrome and 21 general condition evaluation indicators(stress degree of rats,hair moisture degree,stool frequency,etc.),16 individual surface characteristic indicators(systolic blood pressure,diastolic blood pressure,heart rate,etc.),4 serum biochemical indicators(triglyceride,total cholesterol,high-density lipoprotein cholesterol and low-density lipoprotein cholesterol)and 15 behavioral indicators(total distance of open field experiment,total distance of elevated cross maze,etc.)were measured to evaluate the model by orthogonal partial least squares discriminant analysis(OPLS-DA).Results:At the 8th week,the blood pressure and macroscopic body surface characteristics of rats reached the standard.At the 20th week,there were significant differences in behavioral indicators.The total score of general condition evaluation,the 10 body surface characteristics indicators(systolic blood pressure,diastolic blood pressure,average arterial pressure,heart rate,body mass,food intake,water consumption,stool water content,face temperature,claw color r value)and the 7 behavioral indicators(pain threshold value,open field experiment:total distance and peripheral distance,cross maze experiment:total distance,times of entering the center,closed arm distance and times of entering the closed arm)were screened by the importance projection discrimination of model variables,and the values were greater than one.Conclusions:WKY rats fed with high-fat and high-salt diet for 20 weeks can be used as an animal model of high-normal blood pressure with excessive phlegm-dampness
