摘 要:基于Hg^(2+)对明胶-金纳米粒子(G-AuNPs)类过氧化物酶活性的增强作用,构筑了一种新型的Hg^(2+)传感器。G-AuNPs具有类氧化物酶活性和类过氧化物酶活性,能够催化O_(2)或H2O_(2)氧化3,3,5,5’-四甲基联苯胺(TMB)生成氧化TMB(oxTMB)的反应,在652 nm处出现紫外可见吸收特征峰。优化条件下,Hg^(2+)能够增强G-AuNPs的类过氧化物酶活性,使吸收信号升高,且升高幅度与Hg^(2+)浓度呈线性关系,在0~15μmol/L和15~35μmol/L范围内的线性方程分别为y=1.314+0.0135x和y=0.704+0.055x,检出限为1.65μmol/L。该传感体系对Hg^(2+)具有良好的选择性,其他常见金属离子不干扰对Hg^(2+)的检测。所设计传感器具有无需修饰、检测时间短、选择性好的优点,有望用于污染水现场Hg^(2+)的检测。A novel Hg^(2+)sensor was constructed using peroxidase mimics based on reinforced gelatin-gold nanoparticles(G-AuNPs).The prepared G-AuNPs exhibits oxidase-and peroxidase-like activities,which can catalyze the oxidation of 3,3,5,5'-tetramethylbenzidine(TMB)by O_2 or H_2O_2 to form oxidized TMB(ox TMB),followed with an UV-vis absorption peak at 652 nm as a characteristic signal.Under the optimal conditions,the addition of Hg^(2+)enhanced the peroxidase-like activity of G-AuNPs,and further raised the absorption signal.The increase level was linearly related to the Hg^(2+)concentration,and the linear equations were y=1.314+0.0135x and y=0.704+0.055x for Hg^(2+)concentrations in the ranges of 0-15μmol/L and 15-35μmol/L,respectively.The detection limit was 1.65μmol/L.The sensing system has favorable selectivity for Hg^(2+),and the other common metal ions showed no interference.The benefits of the proposed sensor include free-modification,short detection time and satisfactory selectivity.Therefore,it owns anticipated application prospect for the on-site detection of Hg^(2+)in polluted water.
