摘 要:目的探究槲皮素对多囊卵巢综合征(PCOS)大鼠的改善作用及其机制。方法54只大鼠给予高脂饲料进行喂养,并连续灌胃来曲唑(1 mg/kg)23 d建立PCOS模型;另取10只纳入空白对照组,给予普通饲料。将造模成功的大鼠随机分为模型组、盐酸二甲双胍组(50 mg/kg)及槲皮素低、高剂量组(15、30 mg/kg),灌胃给予相应药物,连续4周。血糖仪检测大鼠血糖水平,ELISA法检测大鼠空腹胰岛素水平和血清TGF-β_(1)、E_(2)、FSH水平,HE染色计数卵巢内各级卵泡数量,RT-qPCR法检测卵巢组织GLUT4 mRNA表达,Western blot法检测卵巢组织AGEs、RAGE、p-Akt、Akt、p-PI3K、PI3K蛋白表达。结果与模型组比较,各给药组大鼠空腹胰岛素和血糖水平降低(P<0.01),血清TGF-β_(1)水平降低(P<0.01),原始卵泡和初级卵泡数量增加(P<0.01),卵巢组织GLUT4 mRNA表达升高(P<0.05,P<0.01);槲皮素高剂量组、盐酸二甲双胍组大鼠血清E_(2)水平降低(P<0.01),卵巢组织AGEs、RAGE、p-Akt/Akt、p-PI3K/PI3K蛋白表达降低(P<0.05,P<0.01)。结论槲皮素可能通过抑制AGEs/RAGE信号通路减轻胰岛素抵抗,促进卵泡发育,发挥对PCOS大鼠的改善作用。AIM To explore the improvement effects and mechanism of quercetin on polycystic ovary syndrome(PCOS)in rats.METHODS Fifty-four rats were induced into PCOS models with 23 days of consecutive letrozole(1 mg/kg).Administration and high-fat diet feeding,in contrast to another ten rats of the blank control group given ordinary diet.PCOS rat models were randomly divided into the model group,the metformin hydrochloride group(50 mg/kg),and low and high dose quercetin groups(15 and 30 mg/kg).The corresponding drug administration by gavage went on for four weeks.The rats had their blood glucose level detected by blood glucose metry;their fasting insulin level and serum TGF-β_(1),E_(2)and FSH levels detected by ELISA;their ovarian follicles counts at all levels detected by HE staining;their ovarian mRNA expression of GLUT 4 detected by RT-qPCR;and their ovarian protein expressions of AGEs,RAGE,p-Akt,Akt,p-PI3K and PI3K detected by Western blot.RESULTS Compared with the model group,all the group intervened with the drug displayed decreased fasting insulin level and blood glucose level(P<0.01),decreased serum TGF-β_(1) level(P<0.01),increased primordial and primary follicles counts(P<0.01),and increased ovarian GLUT 4 mRNA expression(P<0.05,P<0.01).Groups intervened with metformin hydrochloride or high-dose quercetin displayed decreased serum E_(2)levels(P<0.01),and decreased expressions of ovarian AGEs,RAGE,p-Akt/Akt,and p-PI3K/PI3K proteins(P<0.05,P<0.01).CONCLUSION Quercetin may improve PCOS conditions in rats by reducing insulin resistance,promoting follicular development via inhibition of AGEs/RAGE signaling pathway.
