摘  要:目的构建一种负载纳米铜颗粒的温敏性胶原水凝胶,并探索其应用于腹部开放性海水浸泡伤的救治效果。方法将脱细胞牛心包冻干粉与生物还原法制备的纳米铜混合,采用胃蛋白酶消化,制备纳米铜温敏性胶原水凝胶。健康成年SPF级BALB/c小鼠30只,构建小鼠腹部开放性海水浸泡伤模型,按照随机数字表法分为模型组和治疗组,每组15只。模型组在开放腹腔后给予10 ml 20℃海水浸泡处理30 min,然后逐层关腹;治疗组给予腹腔同样海水浸泡,再予以5 ml纳米铜温敏性胶原水凝胶后,逐层关腹。7 d后处死小鼠,HE染色评价2组小鼠胸腹腔各脏器的损伤情况。结果铜颗粒水合粒径大小为(686.7±134.6)nm,Zeta电位为-24.6 mV,表明生成的纳米铜颗粒为纳米级,具有较大的静电斥力,稳定性好。胶原水凝胶在室温时呈液态,37℃时转变为固态,具有良好的温敏性。加入纳米铜颗粒后,水凝胶的孔隙率和吸水率轻度较少,不影响其溶胀比。与模型组比较,纳米铜温敏性胶原水凝胶能提高治疗组小鼠的存活率和生存状态,减轻海水对腹腔脏器的损伤,并减轻腹部伤口的感染程度。结论纳米铜温敏性胶原水凝胶可以减轻高渗海水对小鼠腹腔脏器组织的损伤,能维护重要脏器功能,并具有抗菌活性。Objective To construct a thermosensitive collagen hydrogel loaded with copper nanoparticles and explore its application in the treatment of open abdominal seawater immersion wound.Methods Acellular bovine pericardium lyophilized powder was mixed with nano copper prepared by biologic reduction method,and nano copper temperature sensitive collagen hydrogel was prepared by pepsin digestion.A total of 30 healthy adult SPF BALB/c mice were used for establishing open abdominal seawater immersion injury models,which were then divided into model group and treatment group according to random number table method,with 15 in each group.The abdomen of the mouse in the model group was opened and immersed with 10 ml seawater at 20℃for 30 min,and the cut was closed layer by layer.The mice in the treatment group were treated with 5 ml nano copper thermosensitive collagen hydrogel on the cuts after those treatments in the model group,and the cuts were closed layer by layer.Seven days later,the animals were sacrificed and HE staining was used to evaluate the injury of the organs in the chest and abdomen.Results The hydrated particle size of copper particles was(686.7±134.6)nm and the Zeta potential was-24.6 mV,indicating that the particles were nano-sized,and had large electrostatic repulsion and good stability.The collagen hydrogel was liquid at room temperature and changed to solid state at 37℃.It had good temperature sensitivity.After adding nano copper particles,the porosity and water absorption of the hydrogel were slightly lowered,while the swelling ratio was not affected.Compared with those in the model group,nano copper thermosensitive collagen hydrogel can improve animal survival rate and survival state,reduce the damage of seawater to abdominal organs,and reduce the degree of infection of abdominal wounds.Conclusion Nano copper thermosensitive collagen hydrogel can reduce the damage of hyperosmotic seawater to abdominal organs,maintain the functions of the vital organs,and has antibacterial activity.
