摘　　要：目的 借助计算机技术揭示百合地黄汤治疗抑郁症(MDD)的主要物质与作用靶点,并对其抗抑郁机制展开分析。方法通过中药系统药理学分析平台(TCMSP)对百合地黄汤的化学构成与作用靶点展开检索,借助相关数据库(Genecards、OMIM),进行同MDD相关基因的查询。采用UniProt数据库查询靶点对应的基因,由此借助Cytoscape 3.2.1完成化合物-基因(靶点)网络、蛋白相互作用(PPI)网络的构建,研究其作用机制。结果 发现该方可能是通过白介素6(IL-6)、雄激素受体(AR)以及雌激素受体(ESR1、ESR2)等核心靶基因释放对MDD的治疗作用。结论 在对MDD进行治疗方面,可能通过作用于30个靶点,调节信号通路,参与炎症反应、细胞凋亡、神经递质调节等过程,发挥治疗抑郁症的作用。Objective To analyze the main active substance and target of Baihe Dihuang decoction in the treatment of depression(MDD) were revealed by computer technology,and its anti-depression mechanism.Methods The chemical composition and target of Baihe Dihuang decoction were searched by traditional Chinese medicine systematic pharmacological analysis platform(TCMSP),and the genes related to MDD were queried by Genecards and OMIM.Cytoscape 3.2.1 was adopted to construct the compound gene(target) network and protein interaction(PPI) network,and investigate the mechanism of the gene corresponding to the target using UniProt database.Results It was found that this prescription may have the therapeutic effect on MDD through the release of core target genes such as interleukin-6(IL-6),androgen receptor(AR) and estrogen receptor(ESR1,ESR2).Conclusion In the treatment of MDD,it may act on 30 targets,regulate signaling pathways,participate in inflammatory response,apoptosis,neurotransmitter regulation and other processes,and play a role in the treatment of depression.
