草鱼生长性状相关微卫星标记的初步筛选

摘  要:以63尾草鱼为样本,利用微卫星分子标记技术初步筛选出与生长性状相关的分子标记。K-S和QQ图分析显示草鱼体长、体高和体重3个主要生长性状均为正态分布。利用SPSS 20.0分析不同基因对草鱼生长性状的影响,共获得13个基因型;同时发现D基因对草鱼生长性状具有负调控作用,而B、C基因对D基因有抑制作用。微卫星标记与草鱼3个主要生长性状关联分析显示,4对引物HLJC107、HLJC111、C2056、HLJC-9均与草鱼不同生长性状相关联,其中,HLJC107与体长极显著相关,与体高、体重显著相关;HLJC111与体长、体重显著相关;HLJC-9与体长、体高极显著相关;C2056仅与体重显著相关。In this paper,63 fish were used as samples to screen the microsatellite molecular markers related to the growth traits of Grass carp.The results of the K-S analysis and the QQ map showed that the growth traits of the body length,the body height,and the body weight are in a normal distribution.The effect of the microsatellite markers on the growth traits was analyzed by SPSS 20.0 and 13 genotypes were obtained.Also,it was found that the D gene had negative regulatory effect on the growth of Grass carp,while the B and C genes could suppress the expression of the D gene.The association analysis of microsatellite markers with growth traits showed that the 4 pair primers of HLJC107,HLJC111,C2056,and HLJC-9 were respectively related to different growth traits of Grass carp.Among them,the HLJC107 locus was extremely related to the body length,the body height,and the body weight.The HLJC111 locus was significantly correlated with the body length and the body weight.The HLJC-9 locus was extremely related to the body length and the body height.The C2056 locus was only related to the body weight.Based on the results,it can be used as candidate loci for applying to molecular markers assisted breeding of Grass carp.
