摘  要:【目的】探究健脾益肾方对顺铂诱导急性肾损伤小鼠的保护作用及机制。【方法】将12只8周龄雄性C57BL/6J小鼠随机分为空白对照组、模型对照组、健脾益肾方干预组,每组4只。健脾益肾方干预组预先给予15.73 g·kg^(-1)·d^(-1)健脾益肾方灌胃6 d,第6天模型对照组和健脾益肾方干预组小鼠腹腔注射顺铂20 mg·kg^(-1)诱导急性肾损伤,空白对照组腹腔注射等体积的生理盐水。造模72 h后处死取材,检测血清肌酐及尿素氮含量,采用超高效液相色谱检测肾组织三磷酸腺苷(ATP)、单磷酸腺苷(AMP)、二磷酸腺苷(ADP)的含量。【结果】(1)与空白对照组比较,模型对照组血清肌酐、尿素氮水平显著增高(P<0.01);健脾益肾方干预组小鼠血清肌酐水平显著低于模型对照组(P<0.01),尿素氮有下降趋势但差异没有统计学意义(P>0.05)。(2)模型对照组小鼠肾组织AMP含量及AMP/ATP比值较空白对照组显著降低(P<0.05或P<0.01),健脾益肾方干预组小鼠肾组织AMP含量以及AMP/ATP比值显著高于模型对照组(P<0.05),而ATP、ADP含量及ADP/ATP比值的3组组间比较,差异无统计学意义(P>0.05)。【结论】健脾益肾方可有效抑制顺铂诱导小鼠急性肾损伤,对肾脏有保护作用,其机制可能与调控AMP、ATP能量代谢有关。Objective To explore the protective effect and mechanism of Jianpi Yishen Prescription on cisplatininduced acute renal injury in mice.Methods Twelve 8-week-old male C57BL/6J mice were randomly divided into the blank control group,the model control group,and the intervention group with Jianpi Yishen Prescription,with 4 mice in each group.The mice in the intervention group were pre-gavaged with 15.73 g·kg^(-1)·d^(-1) of the Jianpi Yishen Prescription for 6 days,and acute kidney injury was induced by intraperitoneal injection of cisplatin 20 mg·kg^(-1) in the model control and Jianpi Yishen Prescription groups on day 6,while the same volume of saline was injected intraperitoneally in the blank control group.After 72 hours of modeling,serum creatinine and urea nitrogen were measured,and the content of adenosine triphosphate(ATP),adenosine monophosphate(AMP)and adenosine diphosphate(ADP)in kidney tissue were measured by ultra performance liquid chromatography.Results(1)Compared with the blank control group,the serum creatinine and urea nitrogen levels were significantly increased in the model control group(P<0.01);the serum creatinine level of mice in the intervention group was significantly lower than that of the model control group(P<0.01),and the urea nitrogen had a decreasing trend but the difference was not statistically significant(P>0.05).(2)The AMP content and AMP/ATP ratio in the kidney tissue of the mice in the model control group were significantly decreased compared with those in the blank control group(P<0.05 or P<0.01),and the AMP content and AMP/ATP ratio in the kidney tissue of the mice in the intervention group of the Jianpi Yishen Prescription were significantly higher than those in the model control group(P<0.05),while there was no significant difference in ATP and ADP contents and ADP/ATP ratio among the three groups.Conclusion Jianpi Yishen Prescription can effectively inhibit cisplatin-induced acute renal injury in mice and has a protective effect on the kidney.The mechanism may be related to
