摘 要:【目的】建立广西铁皮石斛中核苷类成分(尿苷、鸟苷、腺苷)以及多糖(D-葡萄糖、D-甘露糖)高效液相色谱(HPLC)含量测定方法,比较不同种植模式与生长年限广西铁皮石斛这些成分的含量差异。【方法】核苷类成分HPLC含量测定采用Zorbax SB Aq柱(250 mm×4.6 mm,5μm);流动相为水-甲醇,梯度洗脱;流速1.2 m L/min;检测波长260 nm;柱温20℃。根据课题组前期建立的HPLC柱前衍生方法测定广西铁皮石斛中多糖的含量。【结果】所有成分方法学考察各项指标均符合要求。含量测定结果显示:仿野生样品相较于大棚种植样品,核苷类成分含量更高,多糖含量较低;1~2年生仿野生样品相较于2年及以上样品,核苷类成分和多糖含量均较高。【结论】建立的HPLC方法可作为广西铁皮石斛药材中尿苷、鸟苷、腺苷及多糖类成分含量的分析方法,不同生长年限和种植模式对广西铁皮石斛的核苷类成分和多糖均有一定的影响,可为广西铁皮石斛的质量控制提供参考依据。Objective To establish a high performance liquid chromatography(HPLC)analytical method on the determination of 3 nucleosides components(uridine,guanosine and adenosine)and polysaccharides(D-mannose and D-glucose)in Dendrobium officinale from Guangxi province and compare the difference of these components between samples in different cultivation patterns and growth years.Methods The nucleosides were determined by HPLC using a Zorbax SB Aq column(250 mm×4.6 mm,5μm);the mobile phase was water-methanol with gradient elution;the flow rate was 1.2 mL/min;the detection wavelength was 260 nm;the column temperature was 20℃.The method was based on the HPLC pre-column derivatization method established by the group to determine the content of Dendrobium officinale from Guangxi.Results All the components met the requirements of the methodological investigation.The results of the content determination showed that the nucleoside content was higher and the polysaccharide content was lower in the imitation wild samples compared to the greenhouse grown samples;the nucleoside content and polysaccharide content were higher in the 1-2 year-old imitation wild samples compared to the 2 year-old and above samples.Conclusion The established HPLC method can be used as an analytical method for the content of uridine,guanosine,adenosine and polysaccharide components in the herbs of Dendrobium officinale from Guangxi.Different growth years and cultivation patterns have certain effects on the nucleoside components and polysaccharides of Dendrobium officinale from Guangxi,which can provide a reference basis for the quality control of Dendrobium officinale from Guangxi.
