摘 要:目的:比较不同产地干、鲜粉葛中化学成分及对其中葛根素、3′-甲氧基葛根素、葛根素芹菜糖苷、大豆苷、染料木苷和大豆苷元6种异黄酮类成分的含量进行测定。方法:选用Symmetry C 18色谱柱(4.6 mm×250 mm,5μm),甲醇-0.2%冰乙酸水溶液为流动相进行梯度洗脱,紫外检测波长为250 nm。利用“中药色谱指纹图谱相似度评价系统(2.0版)”软件处理色谱数据,建立了粉葛指纹图谱,同时进行相似度评价。另外,采用HPLC对6种成分进行了含量测定。结果:建立了粉葛的指纹图谱,共标定了26个共有峰,指认了9种成分,分别是3′-羟基葛根素、葛根素、3′-甲氧基葛根素、葛根素芹菜糖苷、大豆苷、染料木苷、大豆苷元、染料木素及芒柄花素。对其中6种指认的成分进行含量测定,发现各地葛根素含量最多。各地鲜品葛根素平均含量为4.48 mg/g,各产地含量排名前三依次是:文山>丽江>红河,各产地6种异黄酮总含量排名前三依次是:红河>文山>丽江;不同产地6种异黄酮总含量均是鲜品大于烘干粉葛。结论:不同产地粉葛干、鲜的化学成分较相似,但含量存在明显差异,且鲜品粉葛中6种异黄酮总含量高于干品,可为粉葛资源的相关开发利用提供有效的科学依据。Objective To compare the chemical constituents of dried and fresh different producing area and determine the contents of six isoflavones,including Puerarin,3′-MethoxyPuerarin,Puerarin apioside,Daidzin,Genistin and Daidzein.Methods The Symmetry C 18 column(4.6 mm×250 mm,5μm)was used with methanol-0.2%glacial acetic acid aqueous solution as the mobile phase for gradient elution,and the ultraviolet detection wavelength was 250 nm.The chromatographic data were processed by the software of“Similarity Evaluation System for Chromatographic Fingerprint of Chinese Materia Medica(version 2.0)”,and the fingerprints of Pueraria thomsonii were established and the similarity was evaluated.In addition,the contents of the six components were determined by HPLC.Results HPLC fingerprints of Pueraria thomsonii were established.A total of 26 common peaks were identified,and 9 components were determined,namely 3′-Hydroxy Puerarin,Puerarin,3′-Methoxy Puerarin,Puerarin apioside,Daidzin,Genistin,Daidzein,Genistein and Formononetin.The contents of six identified components were determined,and it was found that the content of puerarin was the highest in all places.The average content of puerarin in fresh samples is 4.48 mg/g,and the top three contents in each producing area are Wenshan>Lijiang>Honghe,and the top three total contents of six isoflavones in each producing area are Honghe>Wenshan>Lijiang.The total contents of six isoflavones from different origins were higher in the fresh products than in the dried Pueraria thomsonii.Conclusion The chemical components of dried and fresh Pueraria thomsonii from different habitats are similar,but there are obvious differences in their contents.In addition,that total content of the six isoflavone in the fresh Pueraria thomsonii is higher than that of the dry Pueraria thomsonii,which can provide effective scientific basis for the related development and utilization of Pueraria thomsonii resources.
