摘  要:植物多糖粗提液中蛋白质的去除常使用大量有机试剂,工艺操作条件苛刻,后续废物难以回收利用。Ⅱ型ZTC1+1天然澄清剂是天然高分子絮凝剂,通过“架桥”效应沉降大分子杂质,处理手段方便快捷,无有害气体产生。陈皮中多糖成分含量丰富,但受制于复杂的纯化工艺,未能得以进一步研究和推广。采用天然澄清剂去除陈皮粗多糖溶液中的蛋白质,通过控制澄清剂添加比例、温度和时间来设计正交实验,以蛋白质清除率和多糖保留率作为指标,确定最佳纯化条件,并与传统Sevag法进行比对验证。结果表明,当最佳工艺条件为澄清剂用量为2%/4%,温度因素水平为60℃,A组分保温时间为2 h,去除蛋白质效果最佳,蛋白质去除率及多糖保留率均优于Sevag试剂。A large number of organic reagents are often used to remove proteins from crude extracts of plant polysaccharides.TypeⅡZTC1+1 natural clarifier is a natural polymer flocculant,settling macromolecular impurities through the“bridging”effect,the treatment means is convenient and quick,with no harmful gas generation.Tangerine peel is rich in polysaccharides,but it cannot be further studied and popularized due to the complicated purification process.In this paper,the natural clarifier was used to remove the protein from the crude polysaccharide of tangerine peel,and the orthogonal experiment was designed by controlling the proportion,temperature,and time of the clarifier.The optimal purification conditions were determined with protein clearance rate and polysaccharide retention rate as indexes,and compared with the traditional Sevag method.The results showed that the optimal process conditions were 2%/4%clarifier dosage,60℃temperature factor level,and 2 h holding time of component A,and the protein removal rate and polysaccharide retention rate were higher than those of the Sevag reagent.
