摘 要:【目的】获取西藏沙棘根瘤内生菌中具有生物学活性的假单胞菌。【方法】采用细菌纯培养方法,从西藏沙棘根瘤中分离假单胞菌,通过细菌染色镜检、生化试验及16S rDNA基因序列分析等方法进行鉴定,通过生长曲线、耐受性、解磷及固氮能力定性筛选等实验对分离菌株生物学特性进行检测。【结果】分离菌株在YMA培养基上呈白色或乳白色,革兰氏阴性菌,杆状,16S rDNA序列比对结果表明分离株与参考假单胞菌属同源性为99%,命名为QY-X8。QY-X8在pH为5~8、氯化钠浓度0%~2%仍能存活,具有较强的抗逆性,溶解无机磷和有机磷的溶磷圈/菌圈均大于2 mm,具有较好的溶磷能力,但不具有固氮能力。【结论】QY-X8菌株具有较强的耐受性和解磷作用,可为研发生物菌肥提供基础材料。【Objective】To obtain Pseudomonas with biological activity from endophytic bacteria of Hip⁃pophae thibetana root nodules.【Method】The Pseudomonas was isolated by bacterial culture method,and the isolated bacteria were identified by staining microscope,biochemical test and 16S rDNA gene sequence analysis.The biological characteristics of the isolated strains were tested by growth curve,tolerance,qualitative screening for phosphorus solubilizing and nitrogen fixing ability.【Result】The isolated strains were white or milky white on YMA medium,Gram-negative bacteria,rod-shaped,and 16S rDNA sequence alignment showed that the isolated strains had 99%homology with the reference Pseudomonas,named QY-X8.QY-X8 can survive at pH 5~8 and sodium chloride concentration 0%~2%,and has strong stress resistance,and has the ability to dissolve inorganic phosphorus and organic phosphorus simultaneously.The phosphorus dissolving circle/bacteria circle is larger than 2 mm,indicating that it has good phosphorus dissolving ability,but the nitrogen fixing transparent circle can't be detected on the solid medium.【Conclusion】QY-X8 has strong tolerance and dephosphorization,which can provide basic materials for the research and development of biological bacterial fertilizer.
