摘  要:目的 观察逆灸对阿尔茨海默病(Alzheimer’s disease,AD)伴抑郁大鼠血浆心房利钠肽(atrial natriuretic peptide,ANP)及皮质酮(corticosterone,CORT)水平的影响,探讨逆灸防治AD认知障碍伴抑郁的可能作用机制。方法 将雄性SD大鼠随机分为空白组、假手术组、模型组和逆灸组,每组9只。采用双侧海马注射β淀粉样蛋白25-35 (amyloid β25-35,Aβ25-35)的方法复制AD大鼠模型。逆灸组大鼠取“百会”“肝俞”“肾俞”穴施以逆灸。糖水偏好实验评估大鼠情绪状态,Morris水迷宫检测大鼠空间学习记忆能力,电镜观察海马超微结构,ELISA法检测大鼠血浆ANP、CORT浓度。结果 与假手术组比较,模型组大鼠糖水偏好指数明显下降(P<0.01),逃避潜伏期显著延长(P<0.01),平台停留时间与平台穿越次数显著减少(P<0.01);海马神经元细胞损伤明显,胞浆基质溶解,空化;血浆ANP浓度明显降低(P<0.01),CORT浓度显著提高(P<0.01)。与模型组比较,逆灸组大鼠糖水偏好指数明显升高(P<0.01),逃避潜伏期明显缩短(P<0.01),平台停留时间与穿越平台次数明显增加(P<0.05,P<0.01);海马神经元细胞基本正常,突触结构清晰;血浆ANP浓度升高(P<0.01),CORT浓度降低(P<0.01)。结论 逆灸可以有效减缓AD大鼠抑郁样行为的发生发展,上调大鼠血浆ANP表达、降低CORT水平。调节下丘脑-垂体-肾上腺轴功能可能是其防治阿尔茨海默病引发的抑郁样行为的重要机制之一。Objective To observe the effects of pre-moxibustion on the levels of ANP and CORT in plasma of Alzheimer's disease(AD)rats,and to explore the possible mechanism of pre-moxibustion in the prevention and treatment of AD with cognitive impairment.Methods Male SD rats were randomly divided into blank group,sham operation group,model group,and pre-moxibustion group(9 rats/group).The model was established by bilaterally hippocampal injection of Aβ25-35.In the pre-moxibustion group,pre-moxibustion was applied to"Baihui""Gansu"and"Shenshu".Sugar water preference experiment was used to evaluate the emotional state of rats,Morris water maze was used to detect the spatial learning and memory ability of rats,electron microscope was used to observe the ultrastructure of the hippocampus,ELISA was used to detect the concentration of plasma ANP and CORT.Results Compared with the sham surgery group,the model group showed a decrease in glucose preference index(P<0.01),prolonged escape latency(P<0.01),and decreased plat form stay time and number of crossing platforms(P<0.01),the damage of hippocampal neurons was significant,with dissolution and cavitation of the cytoplasmic matrix,the concentration of ANP in plasma significantly decreased(P<0.01),while the concentration of CORT significantly increased(P<0.01).Compared with the model group,the pre-moxibustion group showed an increase in the sugar preference index(P<0.01),a shortened escape latency(P<0.01),an increase in platform stay time and the number of crossing platforms(P<0.05,P<0.01),hippocampal neurons were basically normal,and synapses could be seen,the plasma ANP concentration increased(P<0.01),while the CORT concentration decreased(P<0.01).Conclusion Pre-moxibustion can effectively slow down the occurrence and development of depression like behavior in AD rats,up-regulate the expression of ANP,reduce the level of CORT in plasma,and regulate the activity of hypothalamic-pituitary-adrenal(HPA)axis,which may be one of the important mechanisms for its prevention and treatment
