摘  要:[目的]了解烟草白粉病对秦烟99 DNA甲基化水平的影响,探明其DNA甲基化在胁迫条件下对烟草的调控作用。[方法]结合聚丙烯酰胺凝胶电泳检测技术,对不同程度感染烟草白粉病秦烟99的基因组DNA甲基化水平进行MSAP分析。[结果]DNA甲基化在白粉病胁迫条件下调控烟草生长,秦烟99感染白粉病程度越高,其DNA甲基化水平越高。[结论]该试验通过研究感染不同程度烟草白粉病的秦烟99间的表观遗传多样性,为探明秦烟99生态适应性获得的表观遗传机制提供理论依据。该试验结果丰富了烟草基因组甲基化方面的研究内容,为今后研究烟草表观遗传学提供一定的参考。[Objective]To understand the influence of Erysiphe cichoracearum DC.on DNA methylation level of Qinyan 99 seedlings and explore the regulation of DNA methylation on Nicotiana tabacum under stress.[Method]The genomic DNA methylation level of Qinyan 99 seedlings with different infection degrees of Erysiphe cichoracearum DC.was analyzed MSAP by using polyacrylamide gel electrophoresis technique.[Result]The higher the degree of Erysiphe cichoracearum DC.infection,the higher the DNA methylation level of Qinyan 99 seedlings.[Conclusion]In this study,the epigenetic diversity of Qinyan 99 infected with different degrees of tobacco powder virus was studied to provide a theoretical basis for exploring the epigenetic mechanism of Qinyan 99’s ecological adaptation.The results of this experiment enrich the research content of Nicotiana tabacum genome methylation,and provide a certain reference for the future study of Nicotiana tabacum epigenetics.
