溃疡性结肠炎患者血清lncRNA Mirt2表达与Th9/Treg细胞失衡的关系

摘　　要：目的探讨溃疡性结肠炎(ulcerative colitis,UC)患者血清lncRNA Mirt2表达与Th9/Treg细胞失衡的关系。方法选取我院97例UC患者(UC组)和90名健康志愿者(对照组),采集外周血,并记录临床资料。利用实时荧光定量PCR技术检测血清lncRNA Mirt2水平,采用酶联免疫吸附法检测血清IL-9、TGF-β、IL-4水平,流式细胞仪检测外周血Th9、Treg细胞。分析血清lncRNA Mirt2表达水平与UC患者的临床参数、Th9/Treg及IL-9、TGF-β、IL-4间的相关性。结果UC组血清lncRNA Mirt2表达水平(0.75±0.16)显著低于对照组(1.13±0.10)(P<0.05)。病变时间≥2年、活动期、临床活动度重度者血清lncRNA Mirt2表达水平显著低于病变时间<2年、缓解期、临床活动度为轻度和中度者(P<0.05)。与对照组相比,UC组Th9、Th9/Treg、IL-9显著增加,Treg、TGF-β、IL-4水平显著降低(P<0.05)。与缓解期相比,活动期Th9、Th9/Treg、IL-9水平升高,而Treg、TGF-β、IL-4水平降低(P<0.05)。UC组血清lncRNA Mirt2表达水平与Th9、Th9/Treg、IL-9呈负相关(r=-0.496、-0.535、-0.517,P均<0.05),与IL-4、Treg、TGF-β呈正相关(r=0.461、0.459、0.476,P均<0.05)。结论UC患者血清lncRNA Mirt2表达水平下调与疾病活动性、活动期严重程度及病程相关,并且与Th9/Treg细胞失衡有关。Objective To investigate the relationship between serum lncRNA Mirt2 expression and Th9/Treg cell imbalance in patients with ulcerative colitis(UC).Methods A total of 97 UC patients(UC group)and 90 healthy volunteers(control group)were selected from our hospital.Peripheral blood was collected and clinical data were recorded.Serum lncRNA Mirt2 levels were detected by real-time quantitative PCR.Serum IL-9,TGF-βand IL-4 levels were detected by enzyme-linked immunosorbent assay.Th9 and Treg cells in peripheral blood were detected by flow cytometry.The correlation between serum lncRNA Mirt2 expression level and clinical parameters,Th9/Treg,IL-9,TGF-βand IL-4 in UC patients was analyzed.Results The expression level of lncRNA Mirt2 in the UC group(0.75±0.16)was greatly lower than that in the control group(1.13±0.10)(P<0.05).The expression level of serum lncRNA Mirt2 in patients with disease duration≥2 years,active stage and severe clinical activity was greatly lower than that in patients with disease duration<2 years,remission stage and mild and moderate clinical activity(P<0.05).Compared with the control group,Th9,Th9/Treg and IL-9 in the UC group were greatly increased,while the levels of Treg,TGF-βand IL-4 were greatly decreased(P<0.05).Compared with the remission period,the levels of Th9,Th9/Treg and IL-9 increased in the active period,while the levels of Treg,TGF-βand IL-4 decreased(P<0.05).The expression level of serum lncRNA Mirt2 in UC group was negatively correlated with Th9,Th9/Treg and IL-9(r=-0.496,-0.535,-0.517,all P<0.05),and was positively correlated with IL-4,Treg and TGF-β(r=0.461,0.459,0.476,all P<0.05).Conclusion The down-regulated expression of serum lncRNA Mirt2 in patients with UC is related to disease activity,severity of active stage and disease course,and is related to the imbalance of Th9/Treg cells.
