摘 要:目的研究人参对脂多糖(Iipopolysaccaride,LPS)诱导的抑郁样行为小鼠肠道菌群的影响。方法将小鼠随机分为空白组(Con)、模型组(Mod)、人参水提物组(GS)、盐酸氟西汀组(Flu);给药7 d后,进行旷场实验和悬尾实验,观察小鼠的行为学表现,并使用16S rDNA技术检测各组小鼠粪便肠道菌群变化及预测抑郁样行为小鼠肠道菌群相关的功能通路。结果与模型组相比,人参水提物组小鼠的活动次数和休息时间明显减少,活动总路程及平均速度显著增加;人参水提物组肠道菌群从门到属水平均发生了变化,部分有益菌的丰度增加,部分有害菌的丰度降低,其中厚壁菌门(Firmicutes)、拟杆菌属(Bacteroides)、毛螺菌属(Lachnospiraceae_FCS020_group)、瘤胃菌属(Ruminococcaceae_UCG_009)、Lachnoclostridium菌属显著增多,均有往正常组回调的趋势;KEGG通路富集分析共发现31条功能通路与抑郁样行为小鼠肠道菌群相关。结论人参具有改善抑郁样行为小鼠肠道菌群失调的作用。Objective To study the effect of ginseng on intestinal flora in LPS-induced depression-like behavior mice.Methods The mice were randomly divided into the control group(Con),the model group(Mod),ginseng aqueous extract group(GS),and fluoxetine hydrochloride group(Flu).Seven days after administration,open-field test and tail suspension test were conducted to observe the behavioral performance of mice,and 16S rDNA sequencing technology was used to detect the change of fecal intestinal flora in mice in each group and predict the functional pathways related to intestinal flora in mice with depressive-like behavior.Results Compared with the model group,the number of activities and rest time of mice in the ginseng aqueous extract group were significantly reduced,and the total activity distance and average velocity were significantly increased.The intestinal flora of the ginseng aqueous extract group changed from phylum to genus level,and the abundance of some beneficial bacteria was increased,while the abundance of some harmful bacteria was decreased.Among them,Firmicutes,Bacteroides,Lachnospiraceae_FCS020,Ruminococcaceae_UCG_009,and Lachnoclostridium increased significantly,which had a tendency to be adjusted back to the normal group.A total of 31 functional pathways were found to be associated with the gut microbiota of mice with depressive behavior through KEGG pathway enrichment analysis.Conclusion Ginseng has the effect of improving intestinal microbiota imbalance in mice with depressive behavior.
